摘 要:研制与评价一种镁离子增强的PETIA法检测血清胱抑素C试剂。在含有聚乙二醇6000和镁离子的pH 7.6的Tris-HC1缓冲液中,胱抑素C与包被有胱抑素C抗体的纳米胶乳微球反应并形成一定浊度,该浊度与样品中胱抑素C浓度成正比。该法线性范围为0~15.00 mg/L,批内和批间精密度( CV)分别为0.54%~1.81%和0.72%~2.23%,平均回收率为99.9%,该法( y)与参考方法(x)具有良好的相关性,线性回归方程和相关系数分别为 y =0.9984 x +0.026,r =0.9984。所述的试剂测定血清胱抑素C,具有操作简便、线性范围宽、灵敏度高的优点,具有推广应用价值。Deve1opment and eva1uation of one magnesium ion enhanced PETIA serum Cystatin C assay reagent. The ana1ysis was based on the fact that the Cystatin C react to the Cystatin C antibody and then antigen-antibody Comp1ex was produced,the turbidity of antigen-antibody comp1ex was direct ratio to the concentration of serum Cystatin C. The method was 1iner in the range of 0~15. 00 mg/L,the within run and between run CV was 0. 54% ~ 1. 81% and 0. 72% ~ 2. 23%, the average recovery was 99. 9%. Comparing this methods( y) with refence method( x),we obtained the regression equations:y=0. 998 4 x +0. 026,r =0. 998 4. This method was simp1e,sensitive and suitab1e for c1inica1 app1ication.
