摘 要:目的探讨白色念珠菌伊曲康唑耐药菌株和敏感菌株分泌型天冬氨酸蛋白酶(SAP)活性与菌株毒力及致病性之间的关系。方法将收集鉴定出的白色念珠菌伊曲康唑耐药株[最小抑菌浓度(MIC)≥1 mg/L]和敏感株(MIC≤0.125 mg/L)各20株分别接种在牛血清白蛋白(BSA)培养基上,计算Pz值,比较2组菌株的SAP活性;制备小鼠人工菌血症模型,观察1个月,比较各组小鼠的死亡率和生存率。结果耐药组和敏感组菌株均可产生特异性透明环,其Pz值分别为0.611±0.015和0.667±0.015,2组菌株SAP活性差异有统计学意义(t=11.935,P<0.01);耐药组小鼠死亡16只,敏感组死亡7只,2组小鼠的死亡率差异有统计学意义(χ2=8.286,P=0.004)。结论白念珠菌伊曲康唑耐药菌株SAP的活性、毒力及对小鼠的致病性均高于敏感株;菌株毒力随着Sap活性的增强而增强;白念珠菌SAP与耐药有一定关系。Objective To investigate the correlation of secreted aspartic proteases (SAP) activity with virulence and pathogenicity of itraconazole-resistant or itraconazole-susceptible Candida albicans strains. Methods The col-lected and identified itraconazole-resistant strains (MIC≥1 mg/L) and itraconazole-susceptible strains (MIC≤0.125 mg/L) of Candida albicans (n=20 each) were inoculated in bovine serum albumin (BSA) medium, respectively. The Sap activity of the two groups of strains was compared by calculating the Pz value. The artificial bacteremia model in mice was prepared. After one-month observation, the mortality and survival rate of mice in each group were compared. Re-sults The strains in both of the itraconazole-resistant group and itraconazole-susceptible group produced specific transparent rings, and the Pz values were 0.611±0.015 and 0.667±0.015 respectively. There was a statistically signifi-cant difference in Sap activity between the two groups (t=11.935, P〈0.01). Sixteen mice died in the itraconazole-resis-tant group and seven died in the itraconazole-susceptible group, with a statistically significant difference in mortality between the two groups (χ^2=8.286, P=0.004). Conclusions The Sap activity, virulence and pathogenicity of itracona-zole-resistant Candida albicans strains are higher than those in the itraconazole-susceptible strains in mice. The viru-lence of the strains is enhanced along with the increasing Sap activity. There is a certain correlation between Sap and drug resistance of Candida albicans.
