摘  要:目的:探讨牛角地黄汤对ITP大鼠血清细胞因子IL-10、TGF-β1及血象、骨髓象的影响。方法:将30只SD大鼠随机分成实验组、模型组和正常大鼠组,每组各10只。实验组和模型组SD大鼠用APS法建立ITP大鼠模型。于造模第7天实验组ITP大鼠予牛角地黄汤干预,模型组ITP大鼠和正常大鼠组大鼠予等体积的生理盐水干预。4周后处死各组大鼠,用ELISA检测各组大鼠血清细胞因子IL-10、TGF-β1浓度,全自动血细胞分析仪检测外周血WBC、HGB及PLT三系变化,骨髓HE染色观察巨核细胞数。结果:与正常大鼠组比较,模型组ITP大鼠血清中IL-10、TGF-β1浓度及PLT数减低,差异有统计学意义(P<0.05);骨髓巨核细胞数明显降低(P<0.05)。与模型组ITP大鼠比较,实验组ITP大鼠血清中IL-10、TGF-β1浓度及PLT数上升,骨髓巨核细胞数增多,差异有统计学意义(P<0.05)。结论:牛角地黄汤能使ITP大鼠血清IL-10、TGF-β1浓度上升,促进ITP大鼠外周血PLT数增多,具有免疫调节作用。Objective: To explore the effects of Niujiao Dihuang Decoction on serum cytokines IL-10, TGF-β, hemogram and myelogram of ITP rats. Method: The 30 SD rats were randomly divided into experimental group, model group and normal group, each 10 rats only. The experimental group and the model group SD rats were established the rat model of ITP by APS. At the seventh day of modeling, experimental group was given Niujiao Dibuang Decoction, and model group, normal rats group was given an equal volume of saline. Rats were killed after 4 weeks. The concentration of serum IL-10, TGF-β1 of each rats were detected by ELISA. The changes of peripheral blood WBC, HGB, PLT three series were detected by automatic blood cell analyzer. The number of bone marrow megakaryocytes was observed by HE staining. Results: Compared with normal rats group, the concentration of serum L-10 TGF-β1, platelet number of model group ITP rats decreased, the difference has statistical signif- icance (P〈0.05).The number of bone marrow megakaryocytes decreased significantly (P〈0.05). Compared with model group, the concentration of serum L-10, TGF-β, platelet number and bone marrow megakaryocytes numbers of experimental group ITP rats all increased, the difference has statistical significance (P〈O.05). Conclusion: Niujiao Dihuang Decoction can increase the concen- tration of serum L-10, TGF-β1 of ITP rats, promote peripheral blood platelet number of ITP rats, and it has the effect of im- mune regulatory.
