白头翁汤治疗晚期结直肠癌的药效基因组学研究

摘　　要：目的 研究白头翁汤治疗对湿热内蕴证结直肠癌患者基因表达谱的影响.方法 收集经病理证实的结肠癌患者5例,辨证均属湿热内蕴.白头翁汤治疗21 d,于治疗前及治疗后取血抽提、纯化mRNA,采用Agilent表达谱芯片进行杂交,计算机软件筛选差异表达基因.结果 与治疗前比较,共筛选出159个差异表达基因,其中表达上调38个,下调121个,差异表达最多的基因为与蛋白结合相关的基因,余者包括与代谢信号转导及物质运输等作用有关的基因.结论 白头翁汤可影响湿热内蕴证结直肠癌患者细胞内一系列基因的表达,这些基因可能与其抗肿瘤机制有关.Objective To investigate the effect of Baitouweng decoction on gene expression profile in damp-heat colorectal cancer using gene chip technology.Methods Five damp-heat type patients with colorectal cancer confirmed by pathology were administrated with Baitouweng decoction for 21 days.Before and after treatment,the blood was taken and the total mRNA was extracted and purified.Agilent gene expression profile chip was used to hybridize and the differentially expressed genes were screened.Results Compared with pretreatment,159 differentially expressed genes were found,including 38 genes up-regulated and 121 genes down-regulated.Most of the differentially expressed genes were related with protein binding;others were related with metabolism,signal conduction and material transportation.Conclusion In treatment of damp-heat colorectal cancer patients,Baitouweng decoction influences the expression of a series of genes,which are related to antitumor effects.
