Simultaneous Detection of 40 WADA Prohibited Substances in Human Urine by High Performance Liquid Chromatography-Electrospray Ionization Tandem Mass Spectrometry

摘　　要：运用酸水解-液液萃取及直接液液萃取相结合的方法对尿样进行前处理,建立了同时筛查WADA《禁用清单》中β-阻断剂和麻醉剂,部分刺激剂及β2-激动剂的检测方法。涵盖药物40种,检出限范围分布在0.1~10ng/mL,在低浓度、WADA技术文件要求最低检测浓度（MRPL）和高浓度时相对回收率分布在1.6%~118.6%。解决了实验室原有方法不能达到技术文件MRPL的问题,在快速筛查尿样时效果良好。方法灵敏、简单、经济、快速,结果准确可靠。A high performance liquid chromatography-electrospray ionization tandem mass spec- trometry method was developed for a simultaneous detection of 40 prohibited substances, cov- ered β-blockers and narcotics, some stimulants and β2-agonists which are included in the pro- hibited list issued by the World Anti-Doping Agency （WADA）. After the pretreatment with acid hydrolysis, extraction and enrichment procedures, the urine sample was injected to the LC MS/MS. The limits of detection were between 0. 1 - 10 ng/mL according to different sub- stances. The spiked recoveries at low concentration, WADA Minimum Required Performance Levels （MRPL） and high concentration were within the range of 1. 6% to 118. 6%. This method was a simple, fast, effective and reliable solvent to our laboratory and now has been successfully applied to the routine analysis and the External Quality Assessment Scheme set out by WADA.
