摘 要:目的:研究福辛普利(fosinopril)对压力超负荷性心肌肥厚大鼠bcl-2、bax基因表达的影响。方法:将36只Wistar大鼠随机分为A组(假手术组)、B组(腹主动脉缩窄组)、C组(腹主动脉缩窄组+福辛普利给药组)。实验中以左心室重量指数(LVW/BW)作为判断心肌肥厚的指标;心肌细胞bcl-2、bax基因表达的变化用间接免疫荧光标记法比较。心肌细胞bcl-2、bax基因表达用流式细胞仪(Flow Cytometry FCM)测试。结果:B组与A组相比,心肌细胞bcl-2基因表达下降(P<0.01),bax基因表达增多(P<0.01);C组与B组相比,bcl-2基因表达增多(P<0.01),bax基因表达下降(P<0.05)。结论:福辛普利可促进压力超负荷性心肌肥厚大鼠bcl-2基因的表达,抑制bax基因的表达,从而使心肌肥厚发生逆转。Objective:To investgate the effects of fosinopril on the bcl-2 and bax genes of myocardial hypertrophy rat with pressure overload. Methods:36 Wistar rats were randomly divided into three groups,A group ( false operation group ) , B group ( abdominal aorta constriction group ) and C group( abdominal aorta constriction +fosinopril treatment group) . The ratio of left ventricular weight vs body weight(LVW/BW)was regarded as the index of myocardial hypertrophy;Cardiomyocytes bcl-2, bax gene expression changes in comparison with the indirect immunofluorescence,which was teasted by Flow Cytometry ( FCM) . Result:Comparing with A group,B group is decreased obviously in the bcl-2 gene expression (P〈0. 01),and increased in the bax gene expression (P〈0. 01). Comparing with B group,C group is increased obviously in the bcl-2 gene expression (P〈0. 01),and decreased in the bax gene expression ( P〈0 . 05 ) . Conclusion:Fosinopril could increase the expression of bcl-2 gene and decrease the expression of bax gene in the hypertrophy rats with pressure verload.
