靶向卵巢癌CT分子探针的制备及其性能评价

摘  要:目的:制备一种靶向卵巢癌的CT分子探针,并对其制备方式及相关性能进行评估。方法:采用乳化挥发法制备包裹不同容积的PFOB微球,微球表面进行叶酸修饰,构建叶酸受体靶向的CT分子探针。通过光学显微镜、电镜、激光粒度仪观察其表征;16排螺旋CT扫描仪观察其体外CT显影能力;采用激光共聚焦显微镜观察其与人卵巢癌SKOV3细胞的结合情况。多组均数间采用方差分析进行比较,组内两组均数间采用LSD-t检测进行比较。结果所制备的叶酸受体靶向CT分子探针平均粒径为302.67±27.83nm至563.68±47.29nm,粒径随PFOB容积增加而增大,组间粒径差异具有统计学意义(P<0.05);扫描电子显微镜显示其分散性较好,呈规则球形。透射电镜显示其为壳膜结构,内部核心为不透光阴影区。体外寻靶实验显示靶向组分子探针与SKOV3细胞表面大量结合,而普通组及游离叶酸受体干扰组未见明显特异性结合。体外CT实验显示该分子探针呈明显高密度,CT值范围为200-800HU,CT值随PFOB容积增加而递增,组间差异存在统计学意义(P<0.05)。结论:通过该方法可成功制备叶酸受体靶向CT分子探针,综合各项性能检测结果,容积比(PFOB/PLGA=1:2)为制备该探针的较佳条件。该分子探针具备良好的CT增强显影潜力及与SKOV3细胞特异性体外结合能力,有望成为卵巢癌活体靶向增强显影的理想CT分子探针。ObJective To prepare a novel folate-receptor targeted perfluoronoctylbromide probes, evaluating the preparation methods and characterization. Methods The targeted CT probes with different volume ratio of PFOB/PLGA were prepared by emulsion volatilization technique, and the surface of the probes was modified by FoI-PEG-COOH. The size distribution and surface potential were measured by Laser Particle Size Analyzer System.The emulsification of probes in different volume ratio were observed under the light microscope (LM). The morphology and structure were observed by scanning electron microscope (SEM) and transmission electron microscope (TEM). The targetablity of the targeted prob to SKOV3 cell was observed by confocal laser scanning microscope (CLSM), and the mean CT value of the probes was acquired by CT scanner. Results The size distribution of the targeted probes with volume ratio was found tobe about 302.67±27.83 nm to 563.68 ± 47.29 nm, and the surface potential was negative. The size of probes was increased with the higher level of volume ratio. The probes were emulsified well in all volume ratio of LM imaging except the 1:1. The probes were spherical, uniform and well dispersed according to the SEM imaging, and a typical core-shell structure with PFOB encapsulated in the central which demonstrated by TEM. A lot of targeted probes were attached to the SKOV3 cell in targeted group, which were much less in control group and folic acid intervention group. The CT value of all probes was rang from 200-800HU. Conclusions In this study, a novel targeted CT probes was prepared by a relatively simple method. The probe was highly effective targeting in vitro and with strong CT enhanced ability. Neverthenes, the volume ratio of PFOB/PLGA in 1:2 maybe a better choice for synthesis the targeted CT probes. Above all, this targeted CT probes has broad application prospects in CT molecular imaging.
