摘  要:目的评价颈交感神经干离断(TCST)对糖尿病大鼠体液免疫及补体C3的影响。方法健康成年雄性SD大鼠90只,体重280~300g,采用随机数字表法将其分为三组(n=30):正常对照组(C组)、糖尿病组(DM组)和TCST+糖尿病组(TCST组)。DM组和TCST组采用腹腔注射1%链脲佐菌素(STZ)60mg/kg制备糖尿病大鼠模型,TCST组于糖尿病模型制备成功后1周行右侧颈交感神经干离断制备TCST模型,C组和DM组仅分离颈交感神经干,但不离断。分别于颈交感神经干离断前(T0)、离断后1周(T1)、3周(T2)、5周(T3)和7周(T4)时取大鼠采集血样,测血糖并采用ELISA法检测血清免疫球蛋白IgA、IgG、IgM以及补体C3的浓度;处死前称重,解剖分离脾脏组织称重,计算脾脏指数[脾脏重量(mg)/大鼠重量(g)]。结果与C组比较,T0~T4时DM组和TCST组血糖、血清IgA及补体C3明显升高,血清IgG、IgM降低,T2~T4时脾脏重量明显减少、脾脏指数明显降低(P〈0.05)。与DM组比较,TCST组T1~T4时血糖、T2~T4时血清IgA及补体C3水平明显降低,血清IgG、IgM及脾脏指数明显升高,脾脏重量明显增加(P〈0.05)。结论TCST对糖尿病大鼠体液免疫功能及补体系统有一定的调节作用。Objective To investigate the effect of transection of the cervical sympathetic trunk(TCST)on humoral immunity and complement C3 in diabetic rats.Methods Ninety male SpragueDawley rats,weighing 280-300 g,were randomly divided into 3groups(n=30each):control group(group C),diabetes mellitus group(group DM),diabetes mellitus+transection of the cervical sympathetic trunk group(group TCST).Diabetes mellitus was induced by intraperitoneal injection of 1%streptozocin(STZ)60 mg/kg(successful induction of diabetes was defined as blood glucose≥16.7mmol/L)in groups DM and TCST.The right side of cervical sympathetic trunk of the rats in group TCST were broken to prepare the TCST model,while group C and group DM only suffered from the exposure and separation of cervical sympathetic nerve trunk.The concentration of immunoglobin A,G,M(IgA,IgG,IgM)and the complement C3 were recorded by enzyme linked immunosorbent assay(ELISA)before TCST(T0),and on 1st(T1),3rd(T2),5th(T3),7th(T4)week after TCST,the body weight and weight of spleen were measure for spleen index.Results Compared with group C,the level of blood glucose and immunoglobin A,C3 were increased,and the level of immunoglobin G,M and spleen index were reduced at T0-T4(P〈0.05)in groups DM and TCST;Compared with group DM,in group TCST,the level of blood glucose reduced at T1-T4,the level of immunoglobin A,C3 were reduced,the level of immunoglobin G,M was increased,and the level of spleen index was increased at T2-T4(P〈0.05).Conclusion TCST can improve the humoral immunity and complement C3 on diabetic rats.
