摘　　要：目的体外探讨脐血间充质干细胞(UCB-MSCs)对系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMNCs)细胞因子分泌的影响。方法1UCB-MSCs的分离、扩增及鉴定;2健康志愿者及SLE患者PBMNCs的分离。3UCB-MSCs与植物凝集素(PHA)刺激的PBMNCs共培养,ELISA法检测共培养前后各组细胞培养上清液中IL-6、IL-10、IL-2、IL-12、IFN-γ的含量。结果 1UCB-MSCs呈梭形贴壁生长,能在体外连续传代,均表达CD13、CD44、CD166,不表达CD34、CD45;2UCB-MSCs能显著下调SLE患者PBMNCs中IL-10、IL-6的分泌而上调IL-2、IL-12、IFN-γ的分泌。结论 UCB-MSCs在体外能改变SLE患者的Th1/Th2极化状态。OBJECTIVE To explore the effect of umbilical cord blood mesenchymal stem cells on the secretion of ctokinesbyperipheralbloodmononuclearcellsfromsystemiclupuserythematosuspatients.METHODS ①The isolation,expansion and cryopreservation of UC-MSCs;②The isolation,purification and cultivation of PBMNCs from SLE patients and normal person.③UCB-MSCs and PBMNCs stimulated by PHA of SLE patients were coculture, ELISA method was used to detect the content of IL-6,IL-10,IL-2,IL-12 and IFN-γin the cell culture supernatants of eachgroupbeforeandafterco-culture.RESULTS ①UCB-MSCsadheredthebottomoftheflaskandwerefusiform shape,could continuously passage in vitro,express CD13,CD44andCD166 but not CD34 and CD45;②UCB-MSCs could down-regulate the secretion of IL-10 and IL-6 of PBMNCs of SLE patients,and up-regulate the secretion of IL-2,IL-12 and IFN-γ.CONCLUSION UCB-MSCs can change the Th1/Th2 polarization in patients with SLE in vitro.
