摘  要:[目的]筛选库普曼卫矛叶片愈伤组织诱导的适宜条件,为建立完整的库普曼卫矛组织培养体系奠定基础。[方法]以库普曼卫矛叶片为外植体,通过75%酒精+84消毒液+0.1%升汞不同时间消毒处理,寻求外植体最优消毒方法。分别采用MS、1/2MS、WPM 3种类型基本培养基寻求最适宜愈伤组织生长的基本培养基类型。以MS为基本培养基,通过6-BA与IBA不同浓度激素的搭配组合,寻求最适宜愈伤组织生长的培养基类型。[结果]不同消毒处理中尤以0.1%的升汞消毒5 min效果最好;不同基本培养基中尤以MS培养基处理效果最理想;不同激素组合中外植体愈伤组织在MS+6-BA 3.0 mg/L+IBA 0.2 mg/L中生长最好。[结论]在p H 5.8-6.0条件下,外植体采用75%酒精0.5 min+0.1%升汞5 min消毒处理,转入MS+6-BA 3.0 mg/L+IBA 0.2 mg/L+30 g蔗糖+7 g琼脂的培养基中所诱导出的愈伤组织生长最好,效果最显著。[Objective] The optimum condition of Euonymus koopmannii leaf inducing callus medium was selected. It would provide basis for Euonymus koopmannii tissue culture system. [Methods] The leaves were treated as the explants with different concentration combination of 75 % alcohol, 84 disinfectant and 0.1% mercuric. 3 types basic culture medium of MS, 1/2MS,WPM were used to confirm the optimal medium for the growth of callus. Through the adoption of MS as basic medium and the combination of different hormone concentration of 6-BA and IBA, the optimal medium for the growth of callus were founded. [Results] The disinfection with 0.1% mercuric for 5 minutes was proved as the best. MS medium was the best effect compared with other basic culture mediums. The hormone combination of MS+6-BA 3.0 mg/L+IBA 0.2 mg/l, was the best for explants callus. [Conclusion] Under pH 5.8-6.0 the best combined project of disinfection with 75% alcohol for 30 seconds and 0.1% mercuric for 5 minutes, and culture mediums with MS+6-BA 3.0 mg/L+ IBA 0.2 rag/L+30 g sucrose +7 g agar were proved as the best for inducing callus.
