摘  要:目的:研究杨桃果提取物(YTGE)对小鼠急性肝损伤的保护作用。方法:将60只昆明种小鼠随机分为正常对照组,模型对照组,YTGE高剂量(YTGEH)、YTGE中剂量(YTGEM)、YTGE低剂量(YTGEL)组,阳性药联苯双酯组(阳性对照组),每组10只。小鼠经YTGE预处理7d后,采用腹腔注射四氯化碳(CCl4)建立小鼠急性肝损伤模型,用比色分析法测定小鼠血清丙氨酸氨基转移酶(ALT)和天冬氨酸氨基转移酶(AST)的活性;测定小鼠肝匀浆超氧化物歧化酶(SOD)的活性及丙二醛(MDA)的含量;HE染色观察各组小鼠肝细胞损伤程度。结果:与模型对照组比较,YTGE能降低CCl4所致急性肝损伤小鼠血清ALT和AST(P<0.01或P<0.05);升高小鼠肝匀浆SOD的活性,降低MDA的含量(P<0.01或P<0.05);能够减轻肝细胞损伤程度。结论:YTGE对CCl4所致小鼠急性肝损伤有保护作用,其机制可能与其抗氧自由基和抑制脂质过氧化作用有关。Objective. To study the protective effect of YTGE on acute liver injury induced by carbon tetra-chloride(CCl4) in mice. Methods.Kunming mice were randomly divided into normal control group, model group, high-, medium- and low- dose of YTGE groups and biphenyldicarboxylate (BPDC) group. After pretreatment of mice by YTGE for 7 days, CC14 was given intraperitoneally to mice to induced acute liver injury for studying the protective effects of YTGE on liver injury. Colorimetric analysis method was used to test the serum alanineaminotransferase (ALT) and aspartate aminotransferase (AST) activities superoxide dismutase (SOD) activity and malondialdehyde (MDA) content of mouse liver homogenate- liver damage was observed by HE dyeing. Results. YTGE could decrease the levels of serum ALT and AST in acute liv- er injury caused by COl4 ( P〈0.01 or P〈0.05, Compare with model group). Increase the activity of SOD, reduce the content of MDA in liver homogenate ( P〈0.01 or P〈0.05, Compare with model group) and decrease the damage degree of liver cells. Conclusion. The result suggests that YTGE may have protective effects on acute liver injury in mice caused by CCl4. The mechanism may be related to elimination of oxygen free radicals and inhibition of lipid peroxidation.
