摘  要:目的探讨中药糖耐康治疗2型糖尿病的作用机制。方法 44只Wistar大鼠随机分为中药组(n=18),空白组(n=20),西药组(n=6)。中药组给予糖耐康灌胃,制备中药血清;西药组给予吡格列酮灌胃制备西药血清;空白组给予蒸馏水灌胃制备空白血清。用3T3-L1脂肪细胞制备胰岛素抵抗模型,分为模型组、吡格列酮组及糖耐康高、中、低剂量组,并以正常脂肪细胞作为空白组。糖耐康高剂量组予含中药血清的培养液,糖耐抗中剂量组予含1份空白血清及2份中药血清配置的培养液,糖耐康低剂量组予含2份空白血清及1份中药血清的培养液,吡格列酮组予含西药血清的培养液,空白组和模型组给予基础培养液。各组均培养72h后采用ELISA法测各组细胞肿瘤坏死因子-α(TNF-α)含量,免疫荧光法观察各组细胞内TNF-α蛋白表达,RT-PCR法测定细胞内TNF-αmRNA水平。结果模型组脂肪细胞TNF-α蛋白水平较正常组显著升高(P<0.01);各给药组TNF-α蛋白水平均显著低于模型组(P<0.01),且糖耐康高、中剂量组显著低于吡格列酮组(P<0.01)。模型组细胞内有大量TNF-α阳性反应颗粒,糖耐康中剂量组和高剂量组及吡格列酮组TNF-α阳性反应颗粒较模型组明显减少。与模型组比较,糖耐康高剂量组、中剂量组及吡格列酮组TNF-αmRNA表达明显减少,且以糖耐康中剂量组最少。结论糖耐康能够有效改善成熟的3T3-L1脂肪细胞胰岛素抵抗,明显降低胰岛素抵抗3T3-L1脂肪细胞中TNF-α的蛋白及其mRNA表达水平,这可能是其治疗2型糖尿病的作用机制之一。Objective To study the effect and mechanism of Chinese medicine Tangnaikang on type 2 diabetes mellitus. Methods Totally 44 wistar rats were randomly divided into Chinese medicine group (n = 18 ), blank group ( n = 20), and western medicine group ( n = 6). Chinese medicine serum; Perfused western medicine Perfused Chinese medicine group with Tangnaikang to prepare group with Pioglitazone to prepare western medicine serum;Perfused blank group with distilled water to prepare blank serum. Insulin resistance model was made with 3T3-L1 adipocyte, then were divided into model group, Pioglitazone group, Tangnaikang group of high, middle and low dosages group, and normal adipocyte was taken as blank group. Tangnaikang high dosage group was given nutrient solution containing Chinese medicine serum. Tangnaikang middle dosage group was given confected nutrient solution containing one part blank serum and two parts Chinese medicine serum. Low dosage group was given nutrient solution containing two parts blank serum and one part Chinese medicine serum. Pioglitazone group was given nutrient solution containing western medicine serum. Blank and model group were given basic nutrient solution. After cultivating for 72 h, determined content of TNF-α with ELISA, observed the expression of TNF-α protein using the immunofluores- cence method, and determined the level of TNF-α mRNA using RT-PCR method. Results TNF-ot protein level of adipocyte in model group increased remarkably compared with normal group (P 〈0. 01 ) ; TNF-α protein level of all medication groups was lower than that of the model group remarkably (P 〈0. 01 ), and that of middle dosage group was lower than that of Pioglitazone group remarkably (P 〈0.01 ). There Tangnaikang high and were abundant TNF-α positive reaction particle in cells of model group.
