摘 要:目的观察焦炉逸散物职业暴露人群尿液中1-羟基芘浓度水平与P53基因第5、第8外显子损伤的关系。方法选取某焦化厂作业工人239人作为研究对象,高效液相色谱法检测尿液1-羟基芘水平;实时荧光定量PCR扩增人群外周血淋巴细胞P53基因外显子5和外显子8,通过循环阈值推算损伤后扩增效率,间接计算损伤指数。结果炼焦车间和焦油车间作业人员尿液中1-羟基芘浓度高于备煤车间和甲醇车间作业人员尿液1-羟基芘浓度;尿中1-羟基芘校准浓度与P53基因第8外显子损伤指数存在正相关。结论焦炉逸散物职业性接触可致第8外显子损伤突变增加,而与第5外显子损伤突变关系不大。Objective To explore the relationship between urinary 1-hydroxypyrene (1-OHP) concentration of the coke oven workers and the genetic damage of the exon 5, exon 8 of the P53 gene. Methods 239 workers from a coke oven plant were recruited, high performance liquid detection(HPLC)were used to detect the urinary 1-OI-IP concentration, the exon 5 and exon 8 genetic damage index of the P53 gene were evaluate by the real-time PCR technique through relative threshold cycle method. Results The urinary 1- OHP concentration of workers in the cove-oven workshop and coke tar workshop were higher than those in the coal preparation workshop and methanol produce workshop~ adjusted urinary 1-OHP concentration were positive correlated with the P53 exon 8 damage index. Conclusions Coke-oven emissions exposure may increase the P53 exon 8 genetic damage, while with no relation with the exonS.
