摘  要:目的探讨半胱氨酰白三烯受体1(Cys LTsR1)拮抗剂孟鲁司特对肠缺血再灌注(I/R)损伤的保护作用。方法大鼠肠I/R模型,大鼠随机分为假手术组、模型组、孟鲁司特(2 mg·kg-1体重)组,检测各组小肠含水量变化,末端脱氧核苷酸转移酶介导的d UTP缺口末端标记测定法(TUNEL)检测各组小肠黏膜细胞的凋亡,蛋白质印迹法检测各组Cys LTsR1、半胱氨酸天冬氨酸蛋白酶9(caspase 9)和半胱氨酸天冬氨酸蛋白酶8(caspase 8)的表达的变化。结果与模型组相比,孟鲁司特组小肠含水量、肠组织黏膜细胞凋亡、Cys LTR1、caspase 9和caspase 8的表达水平均明显降低(P<0.05)。结论孟鲁司特可以减少肠I/R肠黏膜细胞的凋亡,抑制caspase 8和caspase 9的表达可能是重要的凋亡机制。Objective To explore the role of cysteinyl leukotriene re-ceptor 1 ( cysteinyl leukotrienes receptor , CysLTsR1 ) antagonist monte-lukast protective effect on intestinal ischemia -reperfusion ( I /R) injury in rats.Methods Thirty rats were divided into 3 groups ( sham opera-tion group , model group and test group ( montelukast , 2 mg· kg -1 ) . The tissues near to terminal ileum in rats were extracted to detect the water weight.The apoptosis of intestinal mucosa was determined by ter-minal dexynucleotidyl transferase ( TdT ) -mediated Dutpnick end labe-ling( TUNEL ) .The cysteinyl leukotrienes receptor 1, caspase 9 and caspase 8 were detected by Western blotting .Ressults Compared with model group , intestinal water content , intestinal mucosal cells apoptosis , CysLTR1,caspase 8 and caspase 9 expression levels of small intestine in test group were lower with statistical significance ( P 〈0.05 ) . Conclusion Montelukast reduces apoptosis of intestinal mucosal cells in rats after intestinal I /R, the mechanism may be related to the inhibition of expressions of caspase 8 and caspase 9.
