摘  要:目的检测不同神经系统发育阶段铅暴露对仔鼠海马组织中多聚唾液酸转移酶STX基因m RNA转录水平的影响,探讨铅暴露对仔鼠神经系统发育损伤的可能机制。方法实时荧光定量PCR(real-time q PCR)检测各实验组出生后1 d、3周及6周仔鼠海马组织中STX基因m RNA转录水平。结果正常Wistar大鼠海马组织中STX基因m RNA的转录水平随着仔鼠生长发育逐渐降低;母铅组1日龄仔鼠与对照组相比,STX基因m RNA转录水平明显降低,差异有统计学意义(P<0.05);母铅组3周龄仔鼠、断乳后染铅组6周龄仔鼠分别与其对照组相比,STX基因m RNA转录水平差异无统计学意义(P>0.05);母铅6周龄仔鼠与断乳后6周龄各染铅组、对照组相比,STX基因m RNA转录水平明显升高,差异有统计学意义(P<0.05),各时间点仔鼠1 mmol/L与2 mmol/L染铅组相比,STX基因m RNA转录水平差异无统计学意义(P>0.05)。结论不同神经系统发育阶段,低剂量铅暴露影响仔鼠海马组织中STX基因m RNA转录水平,可能是铅暴露损害神经系统发育的原因。Objective To detection of the transcription level of STX gene mRNA in hippocampus of mice exposed lead in differ- ent neural system development stage. To explore the possible mechanism of the harm of the lead on the different development of nevous system. Methods To test the transcription level of STX gene mRNA in hippocampus of the one day, three weeks and six weeks mice by Real time fluorescence PCR( qRT - PCR). Results With the growth of mice, the transcription of the STX gene mRNA was decreased in hippocampus of normal mice. STX expresses in all groups of one day mice, rats in Pb group had signifi- cantly decrease compared with control group( P 〈 0.05 ). The mice at age of three weeks born by female with lead and the mice at age of six weeks which exposed lead after weaning, the transcription of STX gene mRNA had no significant differences com- pared with control( P 〉 0.05 ). The mice at age of six weeks born by female with lead, the transcription of STX gene mRNA had significantly increase compared with the mice at age of six weeks which exposed lead after weaning and control group (P 〈 0.05 ). In all group of mice, there were no significant differences in the level of gene transcription between 1 mmol/L Pb group and 2 mmol/L Pb group (P 〉 0.05). Conclusion Different neural system development stage, the low exposure of lead was effect on the transcription of STX gene mRNA, which may damage the development of nevous system.
