摘  要:目的对红花中亚精胺衍生物进行分离纯化,并研究其抑制[3H]-5-HT再摄取的作用以及光致异构现象。方法采用中压快速制备色谱、Sephadex-LH20柱色谱、制备型HPLC对亚精胺衍生物进行分离纯化,通过ESI-MS、1H-NMR波谱技术进行结构鉴定。采用经典的大鼠脑突触体[3H]-5-HT再摄取抑制实验方法,初步研究亚精胺衍生物对大鼠脑突触体[3H]-5-HT再摄取的抑制活性。用HPLC方法进行光致异构研究。结果成功分离得到了4个亚精胺衍生物,分别鉴定为N1,N5,N10-(Z)-tri-p-coumaroylspermidine(1),N1,N5-(Z)-N10-(E)-tri-p-coumaroylspermidine(2),N1(E)-N5-(Z)-N10-(E)-tri-pcoumaroylspermidine(3),N1,N5,N10-(E)-tri-p-coumaroylspermidine(4)。亚精胺衍生物2和3对大鼠脑突触体[3H]-5-HT再摄取有较好的抑制作用,而亚精胺衍生物1抑制作用较弱。光致异构研究结果显示4个亚精胺衍生物在日光照射下存在相互转化的光致异构现象,并最终可达到同一个平衡状态。结论亚精胺衍生物具有抑制大鼠脑突触体[3H]-5-HT再摄取的作用,不同构型的亚精胺衍生物对[3H]-5-HT再摄取的抑制作用亦不同。亚精胺衍生物在光照条件下不稳定,存在光致异构现象。Objective To isolate and purify coumaroylspermidines from Carthamus tinctorius L.,and to investigate their inhibition effects on [^3H]-5-HT reuptake and the photoisomerization behavior of coumaroylspermidines. Methods Coumaroyl spermidines were isolated and purified by medium pressure flash chromatography,Sephadex LH-20 column chromatographies and preparative HPLC. The chemical structures were identified by ESI-MS and^1H-NMR. The inhibition effects of coumaroylspermidines on [^3H]-5-HT reuptake in rat brain synaptosomes were studied by classical method. Photoisomerization behaviors of coumaroylspermidines were investigated by HPLC. Results Four coumaroylspermidines were isolated and identified as N1,N5,N10-( Z)-tri-p-coumaroylspermidine(1),N1,N5-( Z)-N10-( E)-tri-p-coumaroylspermidine(2),N1( E)-N5-( Z)-N10-( E)-tri-p-coumaroylspermidine(3),N1,N5,N10-( E)-tri-pcoumaroylspermidine(4). Coumaroylspermidine 2 and 3 showed better inhibition effects on [^3H]-5-HT reuptake in rat brain synaptosomes,while coumaroylspermidine 1 showed weaker inhibition effect. The photoisomerization behavior studies showed that the four coumaroylspermidines transformed into each other under sunlight and finally reached an equilibrium state. Conclusion Coumaroylspermidines could have inhibition effects on 5-HT reuptake and the inhibition effects could be different for different isomers. Coumaroylspermidines are unstable and could show photoisomerization behavior under sunlight.
