摘 要:目的观察饥饿胁迫对小鼠认知功能和大脑海马区前脑啡肽原表达的影响。方法将40只小鼠随机分为对照组和饥饿24 h组(下称饥饿组),Morris水迷宫和八臂迷宫测试小鼠的认知功能。RT-PCR检测大脑海马区前脑啡肽原m RNA表达变化情况。结果饥饿组与对照组比较,1在Morris水迷宫测试中,饥饿组潜伏期明显缩短(P<0.05),通过原平台位置次数和穿越目标停留时间百分增加比(P<0.05);2在八臂迷宫测试中,饥饿组参考记忆错误和工作记忆错误减少(P<0.05);3在RT-PCR实验检测前脑啡肽原实验中,饥饿组小鼠大脑海马区前脑啡肽原表达明显降低(P<0.05)。结论 1饥饿胁迫可增强小鼠的认知功能;2饥饿胁迫通过降低小鼠大脑海马区前脑啡肽原来增强认知功能;3脑肠肽水平的调整可能是"脾藏意主思"功能实现的途径之一。Objective To observe the effect~ of hunger stress on cognition and the expression of proenkephalin in hippocampus of mice. Methods 40 mice were randomly divided into the control group and huger for 24 h group (huger group). The cognitive function of mice was examined by Morris water maze and the 8-arm radial maze test. The mRNA levels of PENK in hippocampus were detected by RT-PCR. Results Compared with the control group, (1) The mean time of escape latencies in hunger group was decreased significantly by Morris water maze test (P〈0.05); (2)In the 8-arm radial maze, the reference memory error and working memory error in hunger group were reduced remarkably (P〈0.05); (3)The mRNA expressions of PENK were reduced significantly in hippocampus (P〈0.05). Conclusion (1)The huger stress could increase the cognitive ability of mice; (2) Starvation could increase the ability of cognition by down-regulating the expression of proenkephalin (PENK) in hippocampus of mice; (3)The functional realization of, The expression of brain-gut peptides may be one of the realization mechanisms of "spleen storing idea and governing thought".
