高效液相色谱法测定中药洗发水中侧柏叶槲皮苷的含量

摘　　要：建立中药洗发水中侧柏叶活性成分含量的HPLC测定方法,色谱条件为以十八烷基键合相硅胶填充剂（150×4.6 mm,5μm）;以甲醇-0.01 mol/L磷酸二氢钾溶液-冰醋酸（40∶60∶1.5）为流动相,流速为1 m L/min;柱温：25℃;进样量为10μL;检测波长为254 nm。以侧柏叶槲皮苷对照品作标准曲线,在0.152~1.52 g/L范围内线性关系良好（r=0.999 5）,加样回收率为93.70%,RSD为1.78%。A HPLC method for the determination of active ingredient Platycladus orientalis （L.） Franco in quercitrin containing in herbal shampoo was established in this paper. The chromatographic conditions were confirmed as following： C 18 bonded silica gel column （150 × 4.6 mm, 5 μm） as analytical column, mixed solution by methanol ： 0.01 mol/L of potassium dihydrogen phosphate water solution ： glacial acetic acid = 40 ： 60 ： 1.5 as mobile phase, 1 mL/min as flow rate and column temperature at 25 ℃. The detection wavelength was 254 nm and injection volume 10 μL. Quercitin was employed as control for the standard curve. The linearity of calibration curve, within the concentration range between 0.152 g/L and 1.52 g/L, was excellent （r=0.999 5） with the recovery of 93.70% and relative standard deviations of 1.78%. The method is simple, sensitive, accurate and reliable. It can be used for the determination of quercitin content in herbal shampoo, providing a valuable standard method for the quality control of herbal shampoo as well as of heral medicine as raw material.
