摘  要:目的比较四唑盐(MTT)比色法和相对增殖度法对4种输液用包装材料细胞毒性检查结果的相关性,评价2种方法在检测药包材体外细胞毒性作用上的优缺点。为建立快速有效的药包材检测新标准提供有力依据。方法 L-929细胞分别采用MTT法和细胞增殖度法对4种包装材料的浸提液进行细胞毒性研究,比较材料对细胞的毒性等级,算出细胞的相对增殖度,分析比较2种方法相关性。结果在同样实验条件下,4种药品包材表现出对L-929细胞不同的细胞毒性。与细胞增殖度法比较,MTT法与细胞增殖法检测结果具有很好的相关性r=0.966(P<0.05)。结论与相对增殖度法相比,MTT比色法由于实际检测所需的细胞量相对较少、试验步骤相对简便、检测周期短,因此具有一定的优越性,值得推荐作为细胞毒性检测方法。OBJECTIVE To compare the current relative growth rate method used in vitro cytotoxicity test of the pharmaceutical packaging materials with MTT method, and calculate the relativity of these two methods. To evaluate advantages and disadvantages of these two methods and give a new way of cytotoxicity test in vitro of pharmaceutical packaging materials. METHODS By using extracts of 4 different pharmaceutical packaging materials, L-929 cells were treated for 2 d and 2, 4, 7 d respectively by the MTT assay and proliferation test. The relative proliferation rate of L-929 cells was caculated, and the toxicity was measured and ranked as 5 grades. The data were statistically analyzed. RESULTS Under the condition of this study, 4 pharmaceutical packaging materials showed different grades of the cytotoxicity. Compared with cell proliferation test, the results of MTT assay showed good relativity (r=0.966, P〈0.05). CONCLUSION Compared with the cell proliferation test, MTT assay need less amount of the cells and shows as a quick, simple and effective method for evaluating in vitro cytotoxicity of pharmaceutical packaging materials. So it is worth using MTT assay as a standard cytotoxicity test in vitro for the pharmaceutical packaging materials.
