摘  要:目的建立RP-HPLC法测定半夏草酸钙针晶含量的方法,比较不同炮制方法对草酸钙针晶含量的影响。方法RP-HPLC法。色谱柱:Symmetry Shield RP18(4.6 mm×250mm,5μm),流动相:磷酸二氢钾-TBA(p H2.0)∶甲醇=92∶8,流速:0.6ml·min-1,检测波长:210nm,柱温:30℃,进样量:5μl。结果半夏炮制品草酸钙针晶含量不同:生半夏2.77%>法半夏1.79%>清半夏0.77%>姜半夏0.44%。结论炮制能降低半夏中草酸钙针晶的含量。高效液相法简单快捷,准确可信,为半夏质量标准的制定奠定基础。Objective To establish a method for determing the content of Pinellia tuber calcium oxalate crystal by RP- HPLC method,make comparison of different processing methods on the content of calcium oxalate crystal. Methods RP- HPLC method.Chromatographic column: Symmetry Shield RP18( 4. 6 × 250 mm,5 μ m),mobile phase: potassium dihydrogen phosphate- TBA( ph2. 0) : methanol = 92: 8,flow rate: 0. 6ml · min- 1,detection wavelength: 210 nm,column temperature: 30 ℃,injection volume: 5 μ l. Results The contents of calcium oxalate crystal is different in Rhizoma pinellia tuber processed products: raw Pinellia ternata 2. 77%> Pinellia tuber 0. 79% >alum processed Pinellia 0. 77% >ginger processed Pinellia 0. 44%. Conclusion-Processing can reduce the content of calcium oxalate crystal in Pinellia ternata. HPLC method is simple and quick,accurate and credible,to lay the foundation for the establishment of pinellia tuber quality standards.
