电针对便秘型肠易激综合征大鼠结肠组织PAR4的调节作用

摘　　要：目的观察电针对C-IBS大鼠结肠中蛋白酶活化受体4(PAR4)表达的影响,探讨PAR4在电针治疗C-IBS的作用及意义。方法将32只SPF级Wistar成年雄性大鼠随机分为C-N组(正常对照组)、C-M组(IBS模型组)、C-DD组(电针大肠俞组)、C-DS组(电针上巨虚组),每组8只,用冰水灌胃法建立C-IBS模型。C-DS组和C-DD组电针治疗7d。取结肠组织,采用实时定量RT-PCR和western blot检测大鼠结肠PAR4 mRNA和蛋白的表达。结果实时定量RT-PCR和western blot结果显示:各组大鼠结肠PAR4 mRNA和蛋白相对表达量存在统计学差异。C-M组结肠组织PAR4 mRNA和蛋白的表达水平明显高于C-N组,C-DD组和C-DS组结肠组织PAR4 mRNA和蛋白的表达水平明显低于C-M组;C-DD组与C-DS组之间结肠组织PAR4 mRNA和蛋白的表达水平表现出明显统计学差异。结论电针可通过调节C-IBS大鼠PAR4的表达发挥治疗作用。Objective To observe effects of electro- acupuncture on constipation- predominant IBS rat models,and expression of PAR4 mRNA and protein in IBS rat models' colon,and to discuss the mechanism of IBS's acupuncture treatment. Methods To set Wistar adult rats as the object of the study and to establish models of irritable bowel syndrome rats. The experiment included normal group,model group,EA Shangjuxu group and EA Dachangshu group. The EA Shangjuxu group and EA Dachangshu group were started to be bounded and given EA once in one days,a total of seven times. After the treatment,collect colon tissues,measure expression levels of PAR4 in rats' colon tissue with real- time fluorescent quantitative PCR and western blot. Results Compared with normal control group,IBS model group rats ' PAR4 mRNA and protein expression levels obviously increases. Compared with IBS model group,the expression levels of PAR4 mRNA and protein in EA Dachangshu group and EA Shangjuxu group have significant decreases. Conclusion Acupuncture can cure constipation- predominant irritable bowel syndrome by adjusting PAR4 mRNA and protein expression in rats' colon tissue.
