摘  要:目的探讨新疆地区维吾尔、汉族CD14基因多态性与变应性鼻炎的关系。方法选取在新疆医科大学第一附属医院耳鼻喉科和变态反应门诊就诊的维吾尔、汉族变应性鼻炎患者300例(病例组)与同期体检中心体检健康者300例(对照组)。应用SNa Pshot SNP分型技术对600例样本进行SNP位点分型,检测CD14基因启动子区的-159位点(rs2569190)。采用病例-对照关联分析方法进行基因型和等位基因频率分析。并将新疆地区维吾尔、汉族该位点多态性与北京、重庆、湖北、广东、台湾、德国、日本、美国、澳大利亚等不同种群进行比较。结果新疆地区维吾尔、汉族变应性鼻炎组与对照组CD14/-159位点基因型和等位基因分布频率差异无统计学意义(P>0.05)。汉族变应性鼻炎组与对照组间基因型和等位基因频率分布差异无统计学意义(P>0.05)。维吾尔族变应性鼻炎组与对照组间基因型和等位基因频率分布差异无统计学意义(P>0.05)。新疆地区人群CD14/-159位点多态性与广东、德国、美国、澳大利亚人群相比有统计学意义,而与其他种群相接近。结论新疆地区维吾尔、汉族都存在CD14-159C/T多态性,都以CT基因型和T等位基因为主,未发现CD14/-159多态性和变应性鼻炎发病之间的关联。Objective To investigate the relationship between the CD14 gene -159C/T polymorphisms and allergic rhinitis(AR) in Xinjiang Uyghur and Hans populations. Methods Three hundred individuals had AR and 300 were healthy controls.We identified the CD14/-159 SNP used SNaPshot technical. The case- control analysis was adopted the frequencies of genotypes and alleles. The distribution of genotype frequen- cies and allele in the Chinese Xinjiang Uyghur and Han population were compared with other different pop- ulations from Beijing, Chongqing, Hubei, Guangdong, Taiwan, Germany, Japan, the United States, and Australia. Results The distribution of genotypes or alleles of CD14gene -159C/T had no differences be- tween the AR group and the control group in both the Xinjiang Uyghur and Hans populations (P 〉0.05). The frequencies of genotypes and alleles were not different between the Han AR and Han control group (P 〉0.05).The frequencies of genotypes and alleles were not different between the Uyghur AR and Uyghur control group (P 〉0.05). The distribution of genotype frequencies and allele of CD14-159C/T in the Chi- nese Xinjiang Uyghur and Han population are quite different from Chinese Guangdong Han populations, American, German and Australian (P 〈0.05). Conclusion The CD14/-159C/T polymorphisms were not different between the Chinese Xinjiang Uyghur and Han population. The major genotype frequencies and allele were both CT and T. No relationship between CD14/-159 SNP and AR was found.
