摘　　要：目的探讨WNT信号通路对类风湿关节炎(RA)大鼠炎性因子的影响。方法 2012年10月-2013年9月在辽宁医学院实验动物中心将关节炎模型建立成功且关节炎指数大于2分的3 0只大鼠随机分为胶原诱导的关节炎模型组,WNT信号通路特异性阻断剂(WIF)低、中、高剂量组。另有6只大鼠为正常对照组。分别予以生理盐水1 ml/d(对照组,模型组)、WIF 5 mg/(kg-1d-1)、10 mg/(kg-1d-1)、15 mg/(kg-1d-1)腹腔注射,44 d后处死。应用单因素方差分析观察应用WNT阻断剂前后各组足趾容积,关节炎指数,IL-6、IL-8m RNA和蛋白表达情况。结果模型组足趾容积,关节炎指数,IL-6、IL-8m RNA和蛋白较对照组明显升高,应用WNT信号通路特异性阻断剂(WIF)后足趾容积,关节炎指数,IL-6、IL-8m RNA和蛋白水平明显下降。结论在RA发病过程炎性因子呈明显高表达状态,而WNT信号通路阻断可以明显抑制炎性因子的释放。Objective To explore the effect of WNT signal pathway in the rheumatoid arthritis. Methods Thirty-six arthritis rat models, which had arthritis index greater than 2 scores, were successfully built and were divided into model groups one and two, low dose of WIF group, middle dose of WIF group and high close of WIF group. Besides, 6 rats were treated as normal control group, and they were given normal sodium 1ml/d, WIF 1, 5, 10 and 15mg/（ kg·d-t） by intraperitoneal injection and were killed after 44 days. Then, we compared the volume of toe and the change of IL-6, IL-8. Results The arthritis of model group developed rapidly, and the arthritis index, the volume of toe increased obviously, and the expression of IL-6 and IL-8 increased. The volumes of toe in low, middle high dose groups of WIF improved obviously （P〈0.05）. The protein and mRNA of IL-6 and IL-8 decreased in WIF groups. Conclusion IL-6 and IL-8 highly express in Arthritis. Blocking WNT signal pathway can inhabit the expression of IL-6 and IL-8, and the therapeutic effect shows close dependent.
