摘  要:目的人工合成绿原酸(chlorogenic acid,CGA)完全抗原并进行鉴定,为制备绿原酸单克隆抗体奠定基础。方法采用活泼酯法,将绿原酸分别与牛血清白蛋白(bovine serum albumin,BSA)和卵白蛋白(ovalbumin,OVA)进行偶联,用紫外扫描法、SDS-PAGE电泳法进行鉴定;间接ELISA法测定免疫小鼠血清抗体效价。结果经紫外扫描法和电泳法检测,CGA与载体蛋白成功偶联,免疫小鼠后获得的多抗血清效价达到1×106。结论成功合成绿原酸完全抗原,可用于绿原酸单克隆抗体的制备。Objective To prepare the monoclonal antibody of chlorogenic acid(CGA),synthesize and identify the artificial complete antigen of CGA.Methods The immunizing antigen(CGA-BSA) and coating antigen(CGA-OVA) were synthesized by active ester method.The characterization of synthesis was identified by UV scanning,SDS-PAGE.The antiserum of immunized mice was tested by indirect ELISA.Results CGA was successfully linked to carrier.Indirect ELISA showed that the antiserum titer of immunized mice were above 1 × 10^6.Conclusions The complete antigen of CGA has been successfully synthesized,which can be used further for preparing monoclonal antibody.
