熟地寄生壮骨方对膝骨关节炎模型大鼠抗炎作用的实验研究

摘　　要：目的观察熟地寄生壮骨方对大鼠棉球肉芽肿及木瓜蛋白酶所致膝骨关节炎(KOA)的影响,探讨其抗炎作用。方法 1采用棉球植入法,间接测量药物干预后肉芽肿干质量抑制率;2采用关节腔注射木瓜蛋白酶法复制右膝骨关节炎模型,用螺旋测微尺测量给药前后各组大鼠膝骨关节肿胀度,观察药物对右膝骨关节炎模型的影响,并取右膝关节组织匀浆上清液测定白细胞介素-1(interleukin-1,IL-1)、白细胞介素-6(interleukin-6,IL-6)含量。结果 1与对照组比较,熟地寄生壮骨方各组大鼠肉芽肿湿、干质量均显著降低(P<0.01)2与空白对照组比较,模型组膝骨关节肿胀度显著增大(P<0.01),IL-1、IL-6含量显著增高(P<0.01),组织病理学改变明显;与模型对照组比较,各给药组大鼠给药前膝骨关节肿胀度均基本一致无明显差异(P>0.05);给药后,熟地寄生壮骨方各组大鼠膝骨关节肿胀度均明显缩小(P<0.05,P<0.01)、且IL-1及IL-6含量均显著降低(P<0.01)。结论熟地寄生壮骨方具有抗炎作用,针对膝骨关节炎模型大鼠亦具有较好的抗炎作用。Objective To observe the anti-inflammatory effect of Shudi Jisheng Zhuanggu decoction（SJZD） on granuloma induced by cotton ball and papain-induced knee osteoarthritis（KOA）. Methods 1 The granuloma rat granuloma model was induced by cotton ball for the observation of the effect of SJZD on granuloma dry weight inhibition rate. 2RatKOA model was established by injection of papain solution into the right knee joint,and the thickness of right knee joint was measured with spiral micrometer before and after administration for the evaluation of the effect of SJZD on swelling degree of joint. Meanwhile,the levels of interleukin 1（IL-1）and interleukin 6（IL-6）in the tissue homogenate supernatant of the right knee were detected. Results 1 SJZD significantly inhibited the formation of cotton-ball-induced granuloma（P〈0.01 compared with the model group）. 2Swelling degree of the joints and the levels of IL-1 and IL-6 were significantly increased in the model group compared to blank control group（P〈0.01）,and the histopathological changes were severe. After treatment,swelling degree of the joint and the levels of IL-1 and IL-6 of SJZD groups were significantly decreased compared to teh model group（P〈0.05 or P〈0.01）. Conclusion SJZD has certain anti-inflammatory effect,in particular on KOA model rats.
