摘  要:目的构建4对小鼠RPS20(ribosomal protein S20,RPS20)miRNA质粒载体,采用RNA干扰的方法干扰CT26细胞RPS20基因,观察干扰后对CT26细胞RPS20 mRNA、蛋白表达及细胞增殖的影响。方法采用荧光定量PCR及Western-Blot检测干扰后RPS20 mRNA及蛋白的表达,采用MTT法和3-H胸腺嘧啶核苷(3-HTdR)法检测干扰后细胞增殖能力的改变。结果 RNA干扰RPS20基因后,CT26细胞RPS20 mRNA表达水平下降,蛋白表达水平降低,肿瘤细胞生长受到一定程度的抑制。结论 CT26细胞RPS20基因的部分缺失,依然可以抑制细胞的正常生长,进一步提示了RPS20在生命活动中的重要作用。Objective To construct 4 pairs of mice RPS20(ribosomal protein S20, RPS20)miRNA interference plasmid vector, and to observe the mRNA expression, protein expression and proliferation of CT26 RPS20 after interference. Methods The expression of RPS20 mRNA was detected by fluorescence quantitative PCR, protein expression of RPS20 was detected by Western blotting method,and cell proliferation was detected by the method of MTT assay and 3-H thymidine(3-H-Td R)assay. Results The expression of RPS20 mRNA and protein was decreased after interference of RPS20 gene in CT26 cells,the tumor cell growth was inhibited to certain degree. Conclusion The partial knockout of RPS20 gene of CT26 cells still can inhibit the normal growth of CT26 cells, suggesting the important role of RPS20 in cell life.
