摘 要:目的探讨调控微小RNAs(miRNAs)成熟通路相关基因与四川汉族人群胃癌易感的相关性。方法纳入207名四川汉族胃癌患者(病例组)和219名性别与年龄和病例组匹配的四川汉族人群(健康对照组)。运用高分辨率熔点曲线法(HRM)做基因分型,分别检测探讨调控mi RNAs成熟通路相关基因的2个SNP位点(DGCR8基因内的rs3757 G/A和AGO1基因内的rs636832A/G)的基因型和等位基因,比较2组人群基因及等位基因频率分布。结果病例组与对照组的基因分布均符合Hardy-Weinberg平衡;2个位点的基因型频率在病例组和对照组之间无明显差异:rs3757G/A为χ2=4.31,P=0.12;rs636832A/G为χ2=0.65,P=0.72。同时,这2个位点的等位基因频率在2组之间亦无明显差异:rs3757G/A为χ2=1.22,P=0.27,OR=1.22,95%CI为0.85-1.77;rs636832A/G为χ2=0.65,P=0.42,OR=0.88,95%CI=0.65为1.19。结论本组数据并未发现调控mi RNA成熟通路的相关基因的rs3757G/A和rs636832A/G位点的基因多态性与四川汉族人群胃癌的易感相关。Objective To investigate the association of genetic variants of two SNP loci( rs3757 G / A and rs636832 A / G loci) in mi RNA-machinery genes with gastric cancer in Sichuan Han population. Methods Two SNP loci( rs3757 G / A in DGCR8 and rs636832 A / G) in AGO1 were genotyped in 207 patients with gastric cancer [the patient group) and 219 controls( with matching age and gender as the control group). The two SNPs were genotyped by high resolution melting( HRM)method on Light Cycler 480 machine in order to compare the genotypic and allele distributions in these two groups. Results The genotypic distributions in both patients and controls were within Hardy-Weinberg equilibrium. Genotypic frequencies of these two SNPs did not show significant difference between patient group and control group( rs3757 G / A: χ^2= 4. 31,P =0. 12; rs636832 A / G: χ^2= 0. 65,P = 0. 72). Meanwhile,the distribution of allele frequency of the two SNPs also failed to show a significant association with gastric cancer( rs3757 G / A: χ^2= 1. 22,P = 0. 27,OR = 1. 22,95% CI =( 0. 85-1. 77); rs636832 A / G: χ^2= 0. 65,P = 0. 42,OR = 0. 88,95% CI = 0. 65-1. 19)]. Conclusion Although the existing data did not support that the polymorphisms of these two loci( rs3757 G / A,rs636832 A / G loci) in mi RNA-machinery genes contribute to the development of gastric cancer in Sichuan Han population,it would be inadequate to draw solid conclusions.The sample volume should be expanded to improve the data in future research.
