摘  要:目的探讨滋养细胞Fas/FasL基因表达异常、外周T细胞亚群变化对胚胎发育的影响以及环孢素A(CsA)在其中的作用。方法雌性CBA/J小鼠、雄性DBA/2小鼠建立妊娠模型。分别在孕第5天开始经阴道灌注给予不同剂量的CsA乙醇溶液(1mg/kg组、5mg/kg组、25mg/kg组),每日1次直至分娩;RT—PCR法检测小鼠绒毛组织滋养细胞FasL和caspase-3基因表达。部分成模小鼠分别于CsA给药7、14天,流式细胞术检测其肝组织匀浆内CD4^+CD25^+T细胞相对含量。结果3组CsA干预孕鼠胚胎吸收率分别为16.99%、13.42%及16.67%,与对照组(18.42%)比较,差异有统计学意义(P〈0.05);FasL和caspase-3基因表达被抑制,与对照组比较差异有差异有统计学意义(P〈0.05)。CsA干预组CD4^+CD25^+T细胞水平明显增加,与自然流产组比较差异有统计学意义(P〈0.05)。结论CsA可抑制FasL以及caspase-3基因的表达,使小鼠CD4^+CD25^+T细胞增多。在一定范围内,CsA剂量越高,抑制作用越强,从而维持正常妊娠,减少流产率。Objective To observe the effects of cyclosporine A (CsA) at different concentrations on the quantities of Fas/FasL mRNA in mice trophoblast cells, the number of peripheral T cell subgroups and their potential relationship with embryonic development. Methods A pregnant model was established using CBA/J female rats and DBA/2 male rats. On Day 5 of pregnancy, different concentrations of CsA in ethanol solution ( 1,5 and 25 mg/kg) were perfused into in the vaginas of these rats once per day until delivery. Then the quantities of FasL and caspase - 3 mRNA in mice trophoblast cells were determined by RT - PCR. Meanwhile, some model mice were administrated with CsA for 7 and 14 days to measure the number of CD4 ^+ CD25^ + T ceils within liver tissue homogenates through flow cytometry. Results The rate of embryo resorption was 16.99% for Group 1 mg/kg, 13.42% for Group 5 mk/kg, and 16.67% for Group 25 mg/kg, which was significantly different from that of the control (18.42% , P 〈0.05 ). The expression of FasL and caspase -3 in three CsA - treated groups was remarkably suppressed at transcriptional level in comparison with the control ( P 〈 0.05). After CsA intervention, an obvious increase was seen in the number of CD4^ + CD25^ +T cells, compared with the natural absorption group. Conclusion CsA can inhibit the expression of FasL and caspase -3 mRNA, resulting in an increased number of CD4 ^+ CD25 ^+ T ceils in mice. In a certain range, the inhibitory effects were enhanced along with the rise of the concentration of CsA. Thus, normal pregnancy is maintained and the rate of abortion will be reduced.
