摘  要:目的观察1型和2型糖尿病模型鼠心脏胶原蛋白及转化生长因子β1(TGF-β1)的变化。方法采用链脲霉素(STZ)诱导的糖尿病大鼠和db/db糖尿病小鼠建立糖尿病模型。Masson染色方法观察心脏胶原含量及病理学改变,评估心肌纤维化程度;免疫组织化学方法观察心脏Ⅰ型胶原、Ⅲ型胶原、Ⅳ型胶原、层粘蛋白和α-平滑肌肌动蛋白(α—SMA)的变化;免疫组织化学和免疫印迹法检测心脏TGF—β1的改变。结果与正常组相比,STZ糖尿病大鼠和db/db糖尿病小鼠心脏组织中均出现明显的纤维化改变,表现为胶原蛋白过度沉积,胶原面积百分比显著增加,Ⅰ型胶原、Ⅲ型胶原、Ⅳ型胶原、层粘蛋白和α—SMA蛋白表达增加(P〈0.05)。同时,TGF—β1蛋白表达较正常组也明显增多(P〈0.05)。结论糖尿病心肌纤维化进程中多种胶原蛋白和TGF—β1的表达增加。Objective To observe the changes of different collagen types and TGF - β1 in the heart of mice with type Ⅰ/Ⅱ diabetes mellitus. Methods A rat model of diabetic cardiomyopathy was established through injection of streptozotocin, while db/db mice were adopted in the current study. The myocardial pathological changes and myocardial collagen content were detected by Masson staining. The levels of collagen types Ⅰ , Ⅲand Ⅳ, laminin and α - SMA were measured by immunohistochemistry. The quantity of TGF - β1 was determined by immunohistochemistry and Western blotting. Results Compared with the normal group, cardiac fibrosis was obviously seen in STZ - induced diabetic rats and db/db mice, especially excessive deposition of collagen, and remarkable increases in the percentage of collagen area and the amounts of collagen types Ⅰ, m and Ⅳ, laminin and α - SMA (P 〈 0.05). Meanwhile, both diabetic rats and db/db mice produced higher levels of TGF- β1 than the normal (P 〈 0.05). Conclusion The increased amounts of various collagen types and TGF - β1 are determined during the development of cardiac fibrosis induced by diabetes.
