摘  要:构建了AO-RB-SDS-GMFX能量转移体系,并利用此体系使用荧光共振能量转移法建立了一种检测吉米沙星(GMFX)的新方法.在pH=7.0的Britton-Robinson(B-R)缓冲溶液及十二烷基硫酸钠(SDS)介质中,以物质的量之比为1.5∶1的吖啶橙(AO)和罗丹明B(RB)分别作为能量给体与能量受体,GMFX的加入可使RB的荧光猝灭,体系的荧光强度的减小值与GMFX的加入量呈线性关系,将此体系用于吉米沙星片剂的测定,结果与高效液相色谱(HPLC)法一致,样品回收率在98.58%~102.16%之间,RSD在0.68%~2.2%之间,检出限为0.025 mg·L-1.此法快速、简便、灵敏、准确.The energy-transfer system of AO-RB-SDS-GMFX was constructed to establish a new method for the determination of gemifloxacin (GMFX) by means of the fluorescence resonance energy transfer (FRET) technique.In the PH 7.0 Britton-Robinson (B-R) buffer solution and sodium dodecyl sulfate (SDS) medium,acridine orange (AO) and rhodamine B (RB) with the molar ratio of 1.5 ∶ 1 are used as the energy donor and energy acceptor,respectively.The studies indicate that the introduction of GMFX leads to the fluorescence quenching of RB and a linear relationship is found between the decrease-values of systemic fluorescence intensity and increase-values of GMFX.This system can be used to determine the GMFX tablets,and its results are consistent with those measured by the high performance liquid chromatography (HPLC) method,where the sample recovery is between 98.58% and 102.16%,RSD is between 0.68% and 2.2 %,and the detection limit is 0.025 mg· L-1.This method is rapid,simple,sensitive and accurate.
