摘  要:目的:研究黄精多糖对小鼠腹腔源性巨噬泡沫细胞(MFC)形成的抑制作用。方法:体外传代培养细胞,以20 mg/L氧化型低密度脂蛋白(ox-LDL)培养细胞以复制MFC模型。试验分为空白对照(常规培养基)组、模型(常规培养基)组与黄精多糖高、中、低浓度(质量浓度分别为80、40、20 mg/L)组,复制模型的同时给药培养,连续72 h。以油红O染色,显微镜下观察细胞形态,计数10个视野内的细胞总数与MFC数并计算MFC形成率;测定细胞中总胆固醇(TC)、游离胆固醇(FC)含量并计算胆固醇酯(EC)量;Western blot法测定CD36、三磷酸腺苷结合转运子(ABCA)1、胆固醇酰基转移酶(ACAT)1蛋白的表达。结果:空白对照组细胞体积未见明显增大,油红O染色阳性细胞数量少。与空白对照组比较,模型组细胞体积增大,镜下可见细胞红油O染色阳性细胞数量增多,MFC形成率升高,TC、FC、EC含量增加,CD36、ACAT1蛋白表达增强,ABCA1蛋白表达减弱,差异有统计学意义(P<0.01)。与模型组比较,黄精多糖高、中、低浓度组细胞接近梭形,油红O染色阳性细胞数量减少,MFC形成率降低,TC、FC、EC含量减少,CD36、ACAT1蛋白表达减弱,ABCA1蛋白表达增强,差异有统计学意义(P<0.01或P<0.05)。结论:黄精多糖可抑制因ox-LDL诱导的MFC的形成。其机制可能与减少巨噬细胞内胆固醇的含量,下调CD36、ACAT1蛋白,并上调ABCA1蛋白表达有关。OBJECTIVE: To study the inhibitory effects of Polygonati Rhizoma polysaccharide on the formation of enterocoelia- derived macrophage foam cell (MFC) in mice. METHODS: Cells were subcultured in vitro. The cells were cultured with 20 mg/L oxidized low density lipoprotein (ox-LDL) to establish the models of MFC. There were blank control group (routine medium), model group (routine medium), and Polygonati Rhizoma polysaccharide high, medium and low concentration groups (mass con- centrations were respectively 80 mg/L, 40 mg/L and 20 mg/L). During the establishment of the models, the drugs were given for consecutive 72 h. Oil red O was used to stain and the cell morphology was observed, the total number of cells and MFC within 10 horizons and the formation rate of MFC was calculated; the contents of total cholesterol (TC) and free cholesterol (FC) in the cells were determined and the amount of cholesteryl ester (EC) was calculated; western blot was adopted to determine the expres- sions of CD36, ABCA1 and ACAT1 proteins. RESULTS: There was no obvious enlargement of cell volume in the blank control group, and the cells positive in the oil red O stain was little. Compared with blank control group, the cells volume in model group was larger; the cells positive in the oil red O stain was increased under the microscope; the formation rate of MFC was higher; the contents of TC, FC and EC were increased; the expressions of CD36 and ACAT1 proteins was increased and the expression of AB- CA1 protein was decreased, with significant difference (P〈0.01). Compared with model group, the cells in Polygonati Rhizoma polysaccharide high, medium and low dose groups were close to fusiform, among which the cells positive in the oil red O stain were decreased; formation rate of MFC was decreased; the contents of TC, FC and EC were decreased; the expressions of CD36 and ACAT1 proteins were decreased and the expression of ABCA1 protein was increased, with significant difference (P〈0.01 or P〈
