摘  要:目的:研究黄连解毒汤对脑缺血模型大鼠丘脑中氨基酸谷氨酸(Glu)与γ-氨基丁酸(GABA)含量的影响。方法:采用线拴法复制大鼠脑缺血模型。48只大鼠随机均分假手术(等容生理盐水)组、模型(等容生理盐水)组、黄连解毒汤药材水提物(简称水提物,800 g/kg)组、黄连解毒汤药材总生物碱(简称总生物碱,44 mg/kg)组、黄连解毒汤药材总黄酮(简称总黄酮,50 mg/kg)组、黄连解毒汤药材总环烯醚萜(简称总环醚萜,80 mg/kg)组,复制模型成功2 h后ig给药,q12 h,连续7 d。采用2-巯基乙醇(OPA)-柱前衍生高效液相荧光检测法测定大鼠丘脑中Glu和GABA含量。结果:与假手术组比较,模型组大鼠丘脑中Glu、GABA含量减少,差异有统计学意义(P<0.01或P<0.05);与模型组比较,水提物组、总生物碱组、总黄酮组、总环烯醚萜组大鼠丘脑中Glu含量增加,总黄酮组大鼠丘脑中GABA含量增加,差异有统计学意义(P<0.01或P<0.05)。结论:黄连解毒汤药材总黄酮可促进脑缺血模型大鼠丘脑中Glu、GABA含量恢复至正常水平,其与黄连解毒汤抗脑缺血、促进脑功能恢复的关系值得深入研究。OBJECTIVE: To study the effects of Huanglian jiedu decoction on the content of glutamic acid (Glu) and γ-amino- butyric acid (GABA) in the thalami of model rats with cerebral ischemia. METHODS: The suture-occluded method was adopted to establish the model rats with cerebral ischemia. 48 rats were randomly divided into sham-operation group (isometric normal sa- line), model group (isometric normal saline), group of aqueous extract from medicinal materials of Huanglian jiedu decoction (hereinafter referred to as aqueous extract, 800 g/kg), group of total alkaloids of thereof (hereinafter referred to as total alkaloids, 44 mg/kg), group of total flavones thereof (hereinafter referred to as total flavones, 50 mg/kg), and group of total iridoids thereof (hereinafter referred to as total iridoids, 80 mg/kg). The drugs were given to the rats 2 h after successful establishment of the mod- els, ig, q12 h, for consecutive 7 d. 2-mercaptoethanol (OPA) pre-column derivatization HPLC with fluorescence detection was em- ployed to determine the contents of Glu and GABA in the thalami of the rats. RESULTS: Compared with sham-operation group, the contents of Glu and GABA in the thalami of the rats in model group were decreased, with significant difference (P〈0.01 or P〈0.05). Compared with model group, the contents of Glu in the thalami of the rats in the aqueous extract group, the total alka- loids group, the total flavones group and the total iridoids group were increased, and the contents of GABA in the thalami of the rats in the total flavones group were increased, with significant difference (P〈0.01 or P〈0.05). CONCLUSIONS: The total flavo- noids of medicinal materials of Huanglian jiedu decoction can return the contents of Glu and GABA to normal levels in the thalami of model rats with cerebral ischemia, and the relationship with the recoveries of anti-cerebral ischemia and promoting brain function of Huanglian jiedu decoction is worthy of further study.
