益肾通络复方含药血清对兔骨髓间充质干细胞体外增殖和成骨分化的影响

摘　　要：目的:观察益肾通络含药血清对骨髓间充质干细胞(BMSCs)增殖和成骨分化的影响。方法:分离BMSCs分为对照组(A组)、低浓度组(B组)、中浓度组(C组)、高浓度组(D组)、阳性对照组(E组),干预后检测细胞增殖和成骨分化情况。结果:CCK-8:干预24 h、72 h后B、C、D组均高于A组,干预120 h后B组高于A、C、D组。ALP活性干预7、14 d后B、C、D组高于A组,但低于E组。结论:益肾通络含药血清能促进BMSCs体外增殖并诱导其成骨分化。Objective： To observe the effect of serum containing Yishen Tongluo decoction on proliferation and osteogenic differentiation of the bone marrow mesenchymal stem cells（BMSCs） in vitro. Methods： To isolate and culture the BMSCs in vitro. The cells were randomly divided into blank control group （ A group） , low-concentration group （ B group）, medium-concentration group （C group）, high-concentration group （D group）, and positive control group （E group）. Observing the proliferation and osteogenic differentiation of BMSCs. Results： CCK-8： Intervention after 24 h,72 h, OD value of B,C,D group were greater than A group; Intervention after 120 h, OD value of B group greater than A,C, D group. ALP activity results： intervention after 7d, 14 d, B, C, D group were higher than A group, but lower than E group. Conclusion： Serum containing Yishen Tongluo decoction can promote the proliferation of BMSCs in vitro and induce the osteogenetic differentiation at the same time.
