清热活血方药对类风湿患者成纤维样滑膜细胞OPG、RANKL、TNF-α及IL-17表达影响分析

摘　　要：目的:分析清热活血方药对类风湿关节炎患者成纤维样滑膜细胞(RA-HFLS)OPG、RANKL、TNF-α、IL-17表达的影响。方法:采用不同剂量的清热活血方药治疗85例类风湿关节炎患者,采用ELISA法检测成纤维样滑膜细胞中的OPG、TNF-α及IL-17浓度,Western blot法检测RANKL蛋白的表达情况。结果:A、B、C 3组与对照组的TNF-α、IL-17水平、RANKL表达比较差异有统计学意义(F=13.872,P=0.003;F=9.831,P=0.011;F=7.392,P=0.039),A、B、C 3组高于对照组,B、C 2组低于A组(P<0.05),C组低于B组;A、B、C 3组与对照组的OPG分泌差异有统计学意义(F=9.856,P=0.008),A、B、C 3组高于对照组,B、C 2组高于A组,C组高于B组。结论:清热活血方药能够抑制RA-HFLS中RANKL、TNF-α及IL-17OPG的表达,促进OPG的分泌。Objective ： To analyze the Qing-re Huoxue Recipe on patients with rheumatoid arthritis fibroblast-like synovial cells （RA-HFLS） of OPG, RANKL, TNF-α, IL-17 expression. Methods： Different doses of heat and activating blood treating 85 cases of rheumatoid arthritis patients , detect fibroblast-like synovial cells OPG by ELISA , and IL-17 concentrations of TNF-α, the expression of RANKL protein detected by Western blot. Results ： A, B, C three groups and the control group TNF-α, IL-17 levels , RANKL expression difference was statistically significant （ F = 13. 872, P = 0.003; F=9.831, P =0.011; F=7.392, P =0.039） , A, B, C three groups higher, B, C groups lower in group A, group C than in group B; A , OPG secretion differences between B, C three groups and the control group was statistically significant （F = 9. 856, P = 0. 008） , A, B, C three groups higher, B, C in two high- group A, group C than in group B. Conclusion： Heat activating blood can inhibit RA-HFLS in RANKL, TNF-α expression and IL-17OPG and promote secretion of OPG.
