健脾益肠散对溃疡性结肠炎大鼠TNF-α、NF-κBp65的影响

摘　　要：目的:探讨健脾益肠散对溃疡性结肠炎(UC)大鼠TNF-α、NF-κBp65的影响。方法:将60只大鼠随机分为正常组、模型组、柳氮磺吡啶组和健脾益肠散高、中、低剂量组各10只;采用2、4、6-三硝基苯磺酸(TNBS)/乙醇法复制UC大鼠模型;给药21 d后,ELISA法测定各组大鼠血清TNF-α水平、免疫组化法检测结肠组织NF-κBp65阳性表达。结果:模型组大鼠血清TNF-α水平和结肠组织NF-κBp65阳性表达均较正常组明显增加(P<0.01);健脾益肠散高剂量组TNF-α水平和高、中剂量组NF-κBp65阳性表达均较模型组降低,差异有统计学意义(P<0.05或P<0.01);健脾益肠散高剂量组上述指标的变化与柳氮磺吡啶组比较差异不显著(P>0.05),但明显优于其低剂量组(P<0.05)。结论:健脾益肠散对UC肠粘膜免疫的保护作用可能与降低外周血TNF-α水平及结肠组织NF-κBp65的阳性表达有关。Objective： To explore the effect of Jianpi Yichang powder（ JP） on TNF- α,NF- κBp65 in ulcerative colitis（ UC） rats. Method： 60 rats were randomly divided into the normal group,model group,sulfasalazine group,JP high,medium and low- dose group of 10 in each. Be used 2,4,6- trinitrobenzene sulfonic acid（ TNBS） / ethanol method copied UC rat model. The level of serum TNF- α and the positive expression of colon tissue NF- κBp65 were respectively detected by ELISA and mmunohistochemistry in each group of rats after gavage 21 d. Result： The serum TNF- α level and colon tissue NF- κBp65 positive expression in model group were significantly higher than that of the normal group（ P〈0.01）.TNF- α level in JP high dose group and the NF- κBp65 positive expression in JP high,middle dose group compared with model group decreased significantly（ P〈0.05 or P〈0.01）. JP high- dose group,the difference of above indexes with sulfasalazine group was not significant（ P〉0.05）,but significantly better than the low- dose group（ P〈0.05）. Conclusion： Protective effects of JP on intestinal mucosal immunity of UC may be related to the decrease of TNF- α level and NF- κBp65 positive expression.
