益气活血复方对mtTFA在慢性心衰大鼠血清中表达上调的实验研究

摘　　要：目的:观察益气活血复方对mt TFA在心力衰竭大鼠中表达的影响。方法:大鼠80只,随机选取20只为正常组,余大鼠经左冠状动脉前降支结扎法配合力竭式游泳、节食等方法造成慢性心衰模型,将造模成功大鼠分为模型组、中药组、西药组,共4组,均以灌胃给药或蒸馏水28 d。利用蛋白免疫印迹法和RT-PCR法分别检测线粒体转录因子A(mitochondrial transcription factor A,mt TFA)的蛋白和基因表达的变化情况,比色法测定高能磷酸盐(adenosine triphosphate,ATP)浓度,ELISA法测定血清脑钠素(brain natriuretic peptide,BNP)的浓度。结果:与正常组相比,模型组及余各组大鼠ATP及mt TFA水平显著降低(P<0.01),BNP含量显著升高(P<0.01);与模型组相比,中药组和西药组ATP及mt TFA明显升高(P<0.01),BNP含量明显降低(P<0.01);西药组与中药组血清中ATP,mt TFA及BNP含量无明显差异(P>0.05)。结论:益气活血中药可显著降低心衰大鼠BNP的含量水平,改善线粒体能量代谢,改善心功能,促进mt TFA生成,可抑制或逆转心室重构的过程,从而有抗心力衰竭的作用。Objective：To observe the effect of Yiqi Huoxue Decoction on the expression of mtTFA in heart failure in rats. Methods：80 rats were randomly selected,20 were normal group, the other rats by left anterior descending coronary ar- tery ligation method with the exhaustive method type swimming, diet to establish chronic heart failure model. The success- ful model rats were divided into model group, Chinese medicine group, western medicine group, a total of 4 groups, all with intragastric administration or distilled water for 28 days. We detected mitochondrial transcription factor A by protein immu- noblotting and RT - PCR （ mitochondrial transcription factor A, mtTFA） changes of the expression of proteins and genes, determination of high - energy phosphate Colorimetry （ adenosine triphosphate, ATP） concentration, the determination of serum brain natriuretic peptide ELISA （ brain natriuretic peptide, BNP） concentration. Results ： Compared with normal group, model group and the other groups ATP and mtTFA levels were significantly decreased （ P 〈 0. 01 ） and the content of BNP increased significantly （P 〈 0.01 ） ; compared with the model group ,Chinese medicine group and Western medi- cine group ATP and mtTFA were increased significantly （ P 〈 0.01 ） and the content of BNP was decreased （ P 〈 0.01 ） ; the western medicine group and Chinese medicine group had no significant difference in serum ATP, mtTFA and BNP con- tent （ P 〉 0.05 ）. Conclusion ： Yiqi Huoxue Decoction can significantly reduce the levels of BNP in heart failure rats, im- prove mitochondrial energy metabolism,improve heart function, promote the formation of mtTFA and can inhibit or reverse remodeling process, and thus play the role of anti heart failure.
