摘  要:目的探讨miR-200a及miR-155在狼疮性肾炎(LN)患者肾组织中的表达及其在LN发病机制中的作用。方法通过实时荧光定量聚合酶链反应测定30例LN患者和15例健康对照组肾组织中miR-200a及miR-155的表达水平。结果 LN患者肾组织中miR-155的表达水平高于健康对照组,miR-200a的表达水平低于健康对照组,差异均具有统计学意义(P<0.01)。LN患者肾组织中miR-200a的表达水平与狼疮评分呈负相关(r=-0.391,P=0.032),miR-200a的表达水平与尿蛋白呈负相关(r=-0.395,P=0.034)。miR-155与狼疮评分呈正相关(r=0.593,P=0.001),miR-155表达水平与尿蛋白呈正相关(r=0.375,P=0.045),miR-155的表达水平与e GFR呈负相关(r=-0.59,P=0.001)。miR-200a的表达水平与e GFR之间无明显相关性(r=0.22,P=0.242)。结论肾组织中miR-155和miR-200a在狼疮性肾炎患者与健康对照组之间的表达水平有差异,且它们的表达水平与蛋白尿、肾功能及狼疮评分均相关。提示LN患者组织中miR-200a与miR-155的表达与LN患者肾功能恶化和狼疮疾病活动程度密切相关,可作为预测LN进展的重要标志物。Objective To investigate the expression of miR-200a and miR-155 in the renal tissue of patients with lupus nephritis (LN), and to analyze its role in the pathogenesis of LN. Methods We quantified the expression level of miR-200a and miR-155 in the renal tissue of 30 patients with LN and 15 healthy controls through real-time PCR. Results Compared with health controls, the miR-155 level in LN patients was higher, and miR-200a level in LN patients was lower, with statistically significant difference (P〈0.001). miR-200a expression was negatively corre-lated with proteinuria (r=-0.395, P=0.034) and SLEDA (r=-0.391, P=0.032). miR-155 expression was positively correlat-ed with proteinuria (r=0.375, P=0.045) and SLEDA (r=0.593, P=0.001), while miR-155 expression was nagatively corre-lated with estimated GFR (r=-0.5, P=0.001). miR-200a expression showed no significant correlation with estimated GFR (r=0.22, P=0.242). Conclusion The miR-155 and miR-200a are differentially expressed in the renal tissue of LN pa-tients and normal controls, and their expressions are closely related to the proteinuria, renal function and SLEDA. miR-155 and miR-200a expression in renal tissue are closely related to the degree of deterioration of renal function and lupus dis-ease activity, which can serve as important targets in the pathogenesis of lupus nephritis.
