摘 要:[目的]制备丹参酮Ⅰ脂质体,并对其包封率、粒径、电位等理化性质进行考察。[方法]采用薄膜分散法丹参酮Ⅰ脂质体,用琼脂糖凝胶柱色谱法和紫外分光光度法测定包封率,用差示扫描量热法检测丹参酮Ⅰ脂质体各组成物质的相变过程。[结果]丹参酮Ⅰ在1.004~6.024μg/m L范围内线性关系良好(r=0.999 9),琼脂糖凝胶柱色谱法能有效分离脂质体和游离药物,加样回收率为(98.23±0.02)%,平均包封率为(92.56±0.39)%,粒径为(90.64±1.21)nm,电位为(-35.37±0.84)m V。[结论]薄膜分散法可用于制备丹参酮Ⅰ脂质体,制备的丹参酮Ⅰ脂质体的包封率高,粒径均一,电位稳定,药物含量和包封率测定方法准确可靠,专属性强。[Objective] To prepare TanshinoneⅠ-loaded Liposomes(TSⅠ-LP) and investigate its entrapment efficiency(EE), particle size, Zeta potential and other physicochemical properties. [Methods] TS Ⅰ-LP were prepared by film dispersion method, and using Sepharose gel and UV spectrophotometry to determine the entrapment efficiency of TS Ⅰ-LP. Differential scanning calorimetry(DSC)thermograms of the liposomal samples were recorded respectively. [Results] A good linear relationship was showed of TSⅠ in the range of1.004 ~6.024 μg/m L(r =0.999 9). Sepharose gel was well in separating the liposomes from free TS Ⅰ. The average recovery rate was(98.23±0.02)%. The EE of TSⅠ-LP was(92.56±0.39)%. The particle size was(90.64±1.21) nm and the Zeta potential was(-35.37±0.84) m V. [Conclusion] The film dispersion method can be used to prepare TSⅠ-LP with a high EE, a narrow size distribution and a stationary potential. The method which determine the content of TSⅠ-LP is reliable and sensitive.
