摘  要:[目的]设计体内体外试验研究环境内分泌干扰物4-硝基苯酚(4-nitrophenol,PNP)对雄性大鼠肾上腺皮质孕酮分泌的影响。[方法]体内试验采用24只SPF级雄性未成年SD大鼠,随机分为4组,连续2周皮下注射(剂量为0、0.1、1.0和10.0 mg·kg-1的PNP)。最后一次注射24 h后称体质量,然后麻醉处死,收集血液及各脏器,以备测定;为进一步确定PNP对肾上腺皮质孕酮分泌的影响,体外试验利用肾上腺皮质细胞原代培养,细胞贴壁后用不同浓度的PNP(0、10-6、10-5和10-4mol·L-1)培养24 h。[结果]体内试验结果显示:0.1和1.0 mg·kg-1PNP处理组大鼠脾脏质量显著低于对照组(P<0.05),但是体质量及其他脏器质量和脏器指数均无显著性差异(P>0.05);10.0 mg·kg-1组大鼠血清睾酮水平显著升高(P<0.05),而孕酮水平显著降低(P<0.05),雌二醇水平差异不显著(P>0.05);光学显微镜组织学检查未发现注射PNP组肾上腺皮质形态异常,但RT-PCR结果显示10.0 mg·kg-1PNP处理大鼠肾上腺中St AR、P450scc和P450c17 mRNA表达显著增加(P<0.05),而3β-HSD mRNA表达显著降低(P<0.05)。体外试验结果显示:与对照组相比,注射PNP组培养液中孕酮水平显著降低(P<0.05)。[结论]4-硝基苯酚可能主要通过影响肾上腺类固醇合成酶的表达来影响肾上腺皮质孕酮的分泌功能。[ Objectives ] To investigate the effect of 4-nitrophenol(PNP) on adrenocortical function of progesterone secretion in male rats. [ Methods] 24 immature SD male rats were divided into four groups and injected subcutaneously PNP(0,0.1,1.0 and 10.0 mg· kg^-1 body weight)for 2 weeks. 24 hours after the final injection, the blood samples were collected to detect serum steroid hormones, and organs were excised and weighed. In addition, we exposed cultured primary adrenal cell to PNP( 0,10^-6, 10^-5 and 10^-4 mol·L^-1) for 24 hours to further validate whether PNP impact adrenal progesterone secretion. [ Results ] The spleen weights significantly decreased in rats treated with 0.1 (P〈0.01)or 1.0 mg·kg^-1 PNP (P〈0.05). The body weight, other organs weight and organ weight index of rats in PNP-treated groups showed no significant difference compared with the control group (P〉 0.05 ). Serum concentrations of testosterone were significantly increased in rats treated with 10.0 mg·kg^-1 PNP (P〈 0.05 ). In contrast, serum concentrations of progesterone were significantly decreased in 10.0 mg·kg^-1 PNP-treated group(P〈0.05). Histological examination did not find any abnormality of the adrenal cortex in PNP-treated groups. However, the results of RT-PCR revealed that the mRNA expressions of StAR,P450scc, and P450c17 significantly increased in rats treated with 10.0 mg·kg^-1 PNP( P〈0.05), but 3B-HSD mRNA expression significantly decreased in 10.0 mg· kg^-1 PNP treated group (P〈0.05). Moreover, progesterone concentrations in adrenal cells were significantly decreased(P〈0.05)in 10^-4mol· L^-1 PNP group compared to the control group. [ Conclusions] These results suggested that PNP disrupted adrenal progesterone secretion probably via affecting expression of steroid synthetase in male rats.
