摘 要:抑制素(INH)主要是通过抑制促卵泡素(FSH)的合成与分泌来调节卵泡发育和精子发生,而INHα是其重要组成亚基。本实验室前期将INHα基因3个RNAi片段(分别命名为B、C和M)整合到小鼠基因组中得到了转基因小鼠;本研究分析这3个RNAi片段在转基因雌鼠中的抑制效果及对繁殖性能的影响。结果表明:B、C和M片段在转基因雌鼠F_3代中遗传率分别为81.3%、51.1%和77.6%,其3周时抑制效率分别为27.5%、96.1%和75.1%;转基因雌鼠F_0代的第1胎产仔数均高于野生型,而F_1、F_2代B和M片段升高,C片段减少,但差异不显著(P<0.05),此外其F_2代3周子鼠卵巢重和体重显著高于野生型(P<0.05)。综上,INH是体内必需的重要调控基因,不能缺少,过多抑制反而会降低其繁殖性能;遗传稳定性可能随着干扰抑制效率的提高而降低。The role of inhibin, transforming growth factor-β (TGF-β) superfamily member, is to regulate follicular development and spermatogenesis through the inhibition of synthesis and secretion of follicle-stimulating hormone. Our previous studies, three RNAi fragments interfering INHc~ gene (named B, C and M) have integrated into mouse genome and got transgenic mice. This study analyzed the interference effects of three segments within the mouse genome and its effects on reproductive performance. The results showed that the genetic rates of B, C and M fragments in Fz generation transgenic female mice were 81.3%, 77.6% and 51.1% and inhibition efficiency of three-week were 27.5%, 96.1% and 75.1%, respectively; On the first litter size, F0 generation of transgenic female mice was higher than wild-type, and F1, F2 generation of B and M segments increased, while C fragment reducted, both have no significant difference, in addition to the ovarian weight and body weight of three-week-age offspring in F2 generation were significantly higher than that of wild-type. In conclusions, INH is an important regulatory gene in vivo which must not be missing, too much inhibition reduces the reproductive performance; genetic stability may decrease with the high interference efficiency.
