摘 要:CYP7A1是催化动物肝脏胆固醇转变成胆汁酸的限速酶。本研究采用PCR-SSCP法和DNA直接测序相结合筛查白羽番鸭CYP7A1基因多态性,分析其与肉质和血清生化指标的相关性。结果表明:在CYP7A1基因外显子2检测到沉默突变c.294.A>G,产生3种基因型从、AG和GG,AG和A分别为优势基因型和优势等位基因,其频率分别为0.5517和0.6107;在外显子3检测到沉默突变c.759.T>A,TT和T分别为优势基因型和优势等位基因,其频率分别为0.6552和0.7931;2个SNP均为中度多态位点,且基因型分布均未偏离Hardy-Weinberg平衡。关联分析显示,c.294.A>G突变对肉质和血清生化指标没有产生显著影响;c.759.T>A突变对血清总胆固醇、总蛋白和白蛋白有显著效应,A为低血清总胆固醇的有利等位基因,可能作为降低番鸭胆固醇的一个标记性辅助选择位点。CYP7A1 (cholesterol 7-alpha hydroxy-lase, CYP7A1)is the rate-limiting enzyme produced the bile acid with cholesterol. The research investigated the polymorphism of CYPTA1 gene by PCR-SSCP methods and DNA se- quencing in Muscovy ducks, analysis on the correlation with meats quality and serum biochemical index. The results indicated that the silent mutation c.294 A 〉 G was detected, and it formed 3 genotype AA, AG and GG, AG and A was dominant genotype and allele respectively, the frequency was 0.5517 and 0.6107 respectively. The silent mutation c.759. T〉A was investigated in exon 3, T]? and T was dominant genotype and allele respectively, the frequency was 0.6552 and 0.7931 respectively, all of 2 SNPs belonged to moderate polymorphism and genotype distribution located in Hardy-Weinberg equilibrium. The correlation results showed that c.294.A〉G mutation have significant effect on meat quality and serum biochemical levels, c.759. T〉A mutation also have significant effect on total cholesterol, total pro- tein and albumin, A was the beneficial allele for low for breeding low cholesterol Muscovy duck. level serum total cholesterol, which may be a marker assisted site
