摘 要:目的分别以FSH和SCF为缓释药物、PLGA为药物载体制备PLGA缓释微球,研究微球体内、外释放过程.方法采用复乳-溶剂挥发法制备微球,培养法测定其体外释放过程;大鼠后腿肌肉注射微球,测定其体内释放情况.结果 FSH-PLGA微球体外突释率17.2%,可释放28 d,体内可释放17 d;FSH-PLGA微球体外突释率8.8%,可释放28 d,体内可释放17 d.结论所制FSH和SCF微球在体内外均可稳定的较长时间的释放药物,验证了FSH和SCF微球的可行性.Objective To prepare PLGA microspheres containing follicle stimulating hormone (FSH) or stem cell factor (SCF) and investigate the effects of sustaining release in vitro and in vivo. Methods Microspheres were prepared by a double-emulsification solvent evaporation method, releasing experiments were performed in vitro by culture method and in vivo by injection of microspheres in the rat hind leg muscle. Results The FSH-PLGA microspheres could release for 28 days and the burst release rate was 17.2% in vitro, release for 17 days in vivo; The SCF-PLGA microspheres could release 28 days and the burst release rate was 8.8% in vitro, release for 17 days in vivo. Conclusion The FSH or SCF microspheres could release for long lime in vitro and in vivo, suggesting the method to prepar FSH or SCF microspheres is feasible.
