摘 要:目的探讨环磷酰胺和白消安制作Wistar大鼠非梗阻性无精子症模型的方法.方法 160只SPF级成年雄性Wistar大鼠随机分为10组,每组16只,第A-C组分别予以白消安20 mg/kg,15 mg/kg,10 mg/kg单次腹腔注射,第D-F组分别予以环磷酰胺200 mg/kg,100 mg/kg,75 mg/kg单次腹腔注射,第G-I组分别以白消安40 mg/kg,20 mg/kg,环磷酰胺200 mg/kg单次灌胃给药,第J组予以生理盐水2.0 m L行单次腹腔注射.用药至大鼠的一个生殖周期后睾丸、附睾组织切片找不到精子和精子细胞即为无精症模型成功.结合用药方式、模型成功率等得出实用的成模方式.结果用药后38 d,存活大鼠睾丸及附睾组织切片结果提示第A,B,G,H组达到无精症模型的标准,即睾丸、附睾组织切片找不到精子和精子细胞,其大鼠存活率分别为18.75%、50.00%、6.25%、12.5%,其余各组大鼠无精症模型效果欠佳.各组成模方式的比较,白消安15 mg/kg单次腹腔注射其造模成功率较高且死亡率较低,是实用的Wistar大鼠无精子症模型制作方式.结论白消安15 mg/kg单次腹腔注射可获得Wistar大鼠非梗阻性无精子症模型,模型成功率为50%.Objective To explore a method of building non-obstructive azoospermia model of Wistar rat with cyclophosphamide and busulfan. Methods 160 adult Wistar rats were randomly divided into 10 groups (n = 16) Rats in group 1-3 were given busulfan 20 respectively. Rats in group 4-6 were intraperitoneal injection respectively. mg/kg , 15 mg/kg, 10 mg/kgby single intraperitoneal given cyclophosphamide 200 mg/kg, 100 Rats in group 7-9 were given busulfan mg/kg, 75 mg/kg injection by single 40 mg/kg , 20 mg/kg , Cyclophosphamide 200 mg/kg by single oral administration respectively, Rats in group 10 were given normal saline 2.0 mL by single intraperitoneal injection. If the sperms and the sperm cells could not be found in the biopsy rat testis and epididymis after a reproductive cycle of rat (38 days), the non-obstructive azoospermia model was successfully established. Results The tissue biopsy of testis and epididymis showed the Azoospermia model was successfully build in group A, B, G and H. Sperm and sperm cells in testis and epididymis tissue were not found, and the survival rates of rats were 18.75%, 50%, 6.25% and 12.5%, respectively. The ideal way to build non-obstructive azoospermia model might be gained by single intraperitoneal injection of Busulfan 15 mg/kg, which had the lowest mortality. Conclusion Single intraperitoneal injection of busulfan 15 mg/kg is an ideal method for building non-obstructive azoospermia model of wistar rats, the success rate is 50%.
