摘  要:目的探讨姜黄素通过Fas/Fas L信号通路对胶质瘤细胞的增殖及凋亡产生影响。方法以人胶质细胞瘤U87细胞为研究对象,采用细胞毒性试验检测姜黄素对U87胶质瘤细胞的细胞毒性作用。四甲基偶氮唑蓝(MTT)比色法、平板克隆实验(colony formation assay)观察姜黄素对胶质瘤增殖能力的影响;流式细胞术(FCM)及Caspase-3活性检测细胞凋亡;流式细胞术检测胶质瘤细胞周期;实时定量PCR、Western blot法检测细胞内Fas、Fas L m RNA和蛋白的表达水平。结果随着姜黄素浓度增加,胶质瘤细胞系的细胞毒性作用也增强(P<0.05),24 h的IC50为14.28μmol/L。流式细胞术检测姜黄素阻滞胶质瘤细胞周期于G2/M期。姜黄素作用胶质瘤后Fas和Fas L在m RNA和蛋白表达水平上升(P<0.05)。姜黄素对胶质瘤细胞的增殖及细胞集落形成有抑制作用,并呈浓度和时间依赖性(P<0.05)。结论姜黄素可能通过上调Fas/Fas L通路抑制胶质瘤细胞增殖并促进凋亡。Objective To explore the influence of curcumin on proliferation and apoptosis of glioma cells via Fas/Fas L signaling pathway. Methods Human glioma cell line U87 cells were used. Cytotoxicity assay was performed to determine the cytotoxicity of curcumin on the U87 glioma cells. Cell proliferation was detected with MTT assay and colony formation assay to estimate the effect of curcumin on proliferation capacity of glioma at different doses and times. The flow cytometry was used to observe cell apoptosis with Caspase-3 activity assay and change of cell cycle was determined by flow cytometry. Real-time quantitative PCR and Western blot assay were performed to investigate the influence of curcumin on the expression of Fas and Fas L m RNA and protein. Results As the concentration of curcumin increased, the cytotoxic effect of curcumin was enhanced on U87 cells(P〈0.05), and the concentration of curcumin for inhibiting cell viability(IC50) was 14.28 μmol/L at 24 h. Curcumin induced the cell cycle arrest at G2/M phase. The expressions of Fas and Fas L were increased after curcumin treatment at both m RNA and protein levels(P〈0.05). Furthermore, the proliferation and colony forming of U87 cells were inhibited by curcumin in a time- and does-dependent manner( P〈0. 05).Conclusion Curcumin can effectively inhibit glioma cell proliferation and induce the apoptosis by raising the expression of Fas/Fas L.
