摘　　要：目的观察2型单纯疱疹病毒(HSV-2)对人永生化角质形成细胞株(Ha Ca T)分泌白细胞介素(IL)-19、IL-20的影响,探讨角质形成细胞(KC)对HSV-2病毒感染初期的免疫防御机制。方法用HSV-2病毒感染Ha Ca T细胞,采用荧光实时定量PCR检测感染后24,48,72 h的IL-19、IL-20 m RNA的表达量。结果经HSV-2病毒感染后Ha Ca T细胞分泌IL-19、IL-20 m RNA的表达量在72 h内逐渐上升,与对照组相比均有统计学意义(P<0.05)。结论 KC具有类淋巴细胞样功能,能分泌IL-19、IL-20。在HSV-2感染KC初期,IL-19、IL-20表达上升,参与HSV-2感染的早期免疫防御反应。Objective To observe the effect of herpes simplex virus-2（HSV-2） on the mRNA expression of IL-19 and IL-20 in human eternal cell lines （HaCaT）,and discuss the mechanism of immune defense of human keratinocyte to HSV-2.Methods HSV-2 was used to infect HaCaT cell,detect the mRNA expression of IL-19 and IL-20 after infection 24 h,48 h and 72 h by RT-PCR.Results In resting state,IL-19 and IL-20 were expressed on HaCaT cell.By the infection of HSV-2,the expression levels of IL-19 and IL-20 mRNA were higher than those of control group （P＜0.05） within 72 hours.Conclusion In the beginning of infection,KC which likes lymphocytes can secrete IL-19 as well as IL-20,and may play an important role in the defense of HSV-2 invasion.
