摘　　要：目的:探讨油橄榄叶提取物(olive leaf extract,OLE)和美沙酮联合应用对海洛因依赖大鼠血浆性激素及卵巢血管内皮生长因子(VEGF)表达的影响.方法:将40只健康大鼠随机分成模型(A)组、OLE治疗(B)组、美沙酮治疗(C)组、OLE+美沙酮治疗(D)组.按递增法采用皮下注射海洛因建立海洛因依赖大鼠模型,造模后药物处理30 d,然后分别用药物进行治疗.采用放射免疫分析法检测血浆催乳素(PRL)、雌二醇(E2)、孕酮(P)、促卵泡生成激素(FSH)和促黄体生成激素(LH).用免疫组织化学法检测卵巢VEGF的表达情况.结果:与A组比较,D组、B组和C组血浆中E2、FSH、P含量显著降低,PRL、LH含量显著升高,卵巢VEGF阳性表达的细胞数量显著下降.其中D组作用显著优于单独治疗组.结论:OLE联合美沙酮对海洛因损害大鼠生殖内分泌后的功能恢复作用优于单独治疗组.Objective ： To explore protection role of olive leaf extract （OLE） and methadone on sexual hormone content in plasma and ovarian vascular endothelial growth factor （VEGF） expression in heroin dependence rats. Methods： Forty rats were randomly divided into model group （A） , group B with OLE, group C with methadone and group D with OLE and methadone. Each group of rats was subcutaneously given heroin with increasing dosage to establish heroin administration. After modeling, the rats were administrated with OLE and methadone for 30 d. The contents of prolaetin （PRL）, estradiol （E2）, progesterone （P）, follicle stimulating hormone （FSH） and luteinizing hormone （LH） in plasma were tested by radioimmunoassay. Immunohistochemical analysis was used to determine VEGF expression in rat ovarian. Results：Compared with group A, groups B, C, D showed that the contents of E2, P and FSH in plasma significantly decreased, and PRL,LH contents increased, and VEGF expression significantly reduced. Moreover, the effects of group A were more powerful than those with single drug treatment. Conclusion： The effect of combined OLE and methadone on reproductive endocrine functional recovery was much better than either of the single treatment after heroin-induced injuries in rats.
