摘  要:目的:建立和确证一种简便、准确的测定大鼠全血中银浓度的分析方法。方法:选取铟(115In)为内标,先用硝酸对全血样品进行微波消解处理,再采用电感耦合等离子体质谱法测定消化液中银离子的浓度。结果:内源性物质不干扰银和内标的测定,全血中银的浓度在5.0~1 000μg·L-1范围内线性关系良好,日内精密度RSD≤3.3%,日间精密度RSD≤3.8%;提取回收率为97.5%~101.9%,基质效应为97.4%~99.4%。应用该法研究了大鼠静脉注射纳米银后银的体内经时变化,结果表明,给药(40 nm)后,大鼠血液中银浓度先迅速下降而后上升,之后再缓慢消除。结论:该方法可为纳米银在生物机体内的评价研究提供分析方法参考。Objective:To develop a simple and accurate analytical method for the determination of silver in rat whole blood. Methods: UsingllSIn as internal standard,whole blood samples were digested by microwave digestion with nitric acid ,followed by ICP-MS analysis to determine the concentration of silver ion. Results:No significant endogenous interferences were detected. The calibration curve was linear within the range of 5.0-1 000 μg · L-1. The intra-day and inter-day precisions were no more than 3.3% and 3.8% , respectively. The extraction recoveries were within 97.5%-101.9% and the matrix effects were within 97.4%-99.4%. The method was applied to the pharmacokinetic study of silver in rats after intravenous administration of silver nanoparticles and the results showed that the silver concentration was rapidly decreased then increased and then slowly eliminated. Conclusion:The method could provide reference for the future work aiming to in vivo assessment of silver nanoparticles.
