摘  要:为研究山羊地方性鼻内腺瘤中DNA甲基转移酶(DNMTs)启动子区甲基化状态及其对转录表达的影响,本研究采用甲基化特异性PCR和荧光定量RT-PCR法检测20例正常鼻内腺组织和24例鼻内腺瘤组织中DNMTs基因启动子甲基化状态及其转录表达水平,分析DNMTs基因甲基化对转录表达的影响。结果显示,24例鼻内腺瘤组织中DNMT1和DNMT3b基因启动子甲基化阳性率分别为20.8%(5/24)和41.7%(10/24),与正常鼻内腺组织(未检测到甲基化)均差异显著(p〈0.05)。20例正常鼻内腺组织中DNMT1和DNMT3b基因相对转录表达量分别为1(0.693~1.444)和0.382(0.503~1.840);5例甲基化和19例非甲基化鼻内腺瘤组织中DNMT1相对转录表达量分别为1.56(0.678~1.474)和1.248(0.820~1.905),两者差异不显著;10例甲基化和14例非甲基化鼻内腺瘤组织中DNMT3b相对转录表达量分别为1.165(0.646~1.548)和0.376(0.230~1.231),两者差异显著(p〈0.05)。而DNMT3a在两组间无差异。以上结果表明,鼻内腺瘤组织中DNMT3b基因甲基化与转录表达相关,推测其对鼻内腺瘤具有促进肿瘤相关基因甲基化的作用,导致肿瘤相关基因失活或激活,最终导致肿瘤发生。To investigate the promoter methylation status and mRNA expression of DNA methyltransferases (DNMTs) genes in enzootic nasal tumor, the methylation status and the mRNA expression level of DNMTs gene in 24 cases of enzootic nasal tumor (ENT) tissues and 20 cases of normal nasopharyngeal epithelial (NNE) tissues were detected by the methylation-specifie PCR(MSP) and real-time RT-PCR. The results showed that the methylation expression rates of DNMT1 and DNMT3b gene in ENT tissues were 20.8% (5/24) and 41.7% (10/24), respectively, whereas, no methylationwas detected in the NNE tissues. In addition, the mRNA relative expression level of DNMT1 and DNMT3b were 1 (0.693 to 1.444) and 0.382 (0.503 to 1.840) in 20 cases of NNE tissues, respectively. The mRNA level of DNMT3a was 0.420 (0.603 to 1.659) in 24 cases of ENT tissues, the mRNA level of DNMT1 was 1.56 (0.678 to 1.474) in 5 cases of methyhted ENT tissues and 1.248 (0.820 to 1.905) in 19 cases of unmethyhted ENT tissues. The mRNA level of DNMT3b was 1.165 (0.646 to 1.548) in 10 cases of methyhted ENT tissues and 0.376 (0.230 to 1.231) in 14 cases of unmethyhted ENT tissues, which was significantly difference between the two groups, but the DNMT1 and DNMT3a had no difference in the two groups. The close correlation between DNMT3b methylation and higher mRNA expression suggested that methylation was an important pathway in theregulation of gene expression. DNMT3b probably make a difference in methylation of tumor related gene, which lead to tumor related gene inactivation or activation and lead to tumorigenesis
