摘 要:为研究副猪嗜血杆菌(H.parasuis)细胞致死膨胀毒素(CDT)各亚基(Cdt A、Cdt B和Cdt C)的相关生物学功能,本研究采用基因芯片技术对以CDT各亚基处理的PK-15细胞基因表达谱进行检测和分析。通过比较各亚基处理细胞与空白对照细胞基因表达谱的检测结果显示,Cdt A、Cdt B和Cdt C作用PK-15细胞后,处理细胞中分别有91个基因、126个基因及135个基因出现差异表达变化。根据芯片结果选取变化较显著的基因进行荧光定量RT-PCR验证,结果与基因芯片检测结果基本一致。基因注释(GO)分析表明,这些蛋白分别参与免疫系统调节、生物调控、代谢过程和定位等过程。本研究结果为进一步研究CDT的致病作用机制奠定了基础。The cytolethal distending toxin (CDT) is a pathogenic factor in some of the Gram-negative bacteria, which is consisted of CdtA, CdtB and CdtC subunits. To investigate the impact of PK-15 cells treated with the individual subunit of CDT in Haemophilus parasuis, the gene expression profiles of the PK-15 cells treated with individual subunit were detected with cDNA microarray technology. The results showed that the differential mRNA expressions of 91, 126 and 135 genes were detected in PK-15 cells treated with CdtA, CdtB and CdtC, respectively. In addition, the real-time PCR test revealed that the expression patterns of the candidate differentially expressed genes were consistent with the results of cDNA microarray. By using gene ontology (GO) analysis, these proteins were found to be involved in immune system process, biologlical regulations, metabolic process and localization. These results laid the foundation for further study on the pathogenesis of CDT in H.parasuis infected pigs.
