跨代染镉对子代大鼠学习记忆的影响

摘  要:目的探讨跨代染镉对子代大鼠学习记忆的影响。方法将36只8周大的SD大鼠随机分为4组:饲料中不添加镉组(1组);饲料中按1 mg/kg加入镉组(2组);饲料中按5 mg/kg加入镉组(3组);饲料中按25 mg/kg加入镉组(4组)。雌雄分开饲养,一月后按照雌雄2∶1的比例合笼,产下的幼鼠同亲代大鼠镉暴露方式和剂量继续接触镉,待幼鼠成长至8周,Morris水迷宫检测大鼠学习记忆能力;石墨炉原子吸收仪测定亲代和子代大鼠肝、肾、脑中镉含量和大脑内锌含量。Western bolt测定海马、纹状体、皮质内SYP-1的表达。结果 2组和3组第3天和第4天水定向航行时间延长,差异有统计学意义(P<0.05)。各组穿越平台与穿越有效区域的次数差异无统计学意义。随着染镉剂量的增加,亲代、子代新生和子代8周大鼠大脑、肝脏和肾镉含量逐渐增高,脑内锌含量逐渐降低。各组亲代大鼠肝脏、肾脏和脑镉的含量与子代新生大鼠中镉含量均呈正相关。2、3、4组与1组比较,海马、纹状体和皮质内SYP-1的表达明显降低,差异有统计学意义(P<0.05)。结论镉对大鼠学习记忆有一定影响。Objective To explore the effect of continuous administration of cadmium from the parental generation to first filial generation on learning and memory in rats. Methods 36 eight week SD rats were randomly divided into four groups: no cadmium in feed group ( group 1 ) , 1 mg/kg cadmium in feed group( group 2) , 5 mg/kg cadmium in feed group( group 3 ) , 25 mg/kg cadmium in feed group (group 4). Male and female rats were fed separately. After one month, in order to obtain pregnant rats two male and one female rats in the same group were placed in one cage. After the offspring was weaned, the same feed as their parents was were given to the offspring until to 8 weeks, learning and memory ability of offspring were determined by Morris test. Contents of zinc in brain, and contents of cadmium in liver, kidney and brain of parent and offspring were determined by graphite furnace atomic absorption spectrometer. Amounts of SYP-1 expression in striatum, hippoeampus and cortex were measured by western bolt. Results Directional navigation times were significantly lengthened on the third day and fourth day in group 2 and 3 ( P 〈 0. 05 ). The times of passing through platform and the effective area were no significantly statistical difference. With the increasing of the cadmium dose, contents of cadmium gradually increased, but contents of zinc decreased in brain in parent, new born and week 8 offspring rats. The contents of cadmium in liver, kidney and brain in parent were positively correlated with that in new born offspring. Compared with group 1, amounts of SYP-1 expression in striatum, hippocampus and cortex significantly decreased in group 2, 3 and 4(P 〈 0. 05). Conclusion cadmium can result in learning and memory damage in rats.
